Objective:Androgen was used to establish amimal model of polycystic ovarian syndrome DHEA was injected by hypodermic.Investigated the effect of sodium ferulate on weight and serum hormone levels and the apoptosis of ovary granulosa cells and the expression of Bcl-2 and Bax by means of rats with polycystic ovarian,Study the the effect of sodium ferulate on the apoptosis of ovary granulosa and its therapeutic effect as well as the possible mechanism.Methods:To develope a model of polycystic ovarian in rats,forty 23-day-old females were divided into model groups and blank groups randomly.The rats in model group were injected with DHEA(6mg per 100 grams body weigh)for up to 20 days and the rats in blank group with oil at the same time.Ten rats were selected randomly.Serum hormone levels and multiple follicular cysts in ovary tissue were used to evaluate the results of model.When more than eighty percent rats in model groups were affirmed to develop polycystic ovarian,the rest rats were divided into treat group and control group.Both group contained ten rats.Sodium ferulate was used for up to 2 weeks by intraperitoneal injection(100mg/kg)in treat group.Meanwhile,0.5ml physiological saline were used in control group and blank group.Observe the weigh change everyday.After sodium ferulate interference finished,abdominal aorta blood collection and immunoradiometric assay were used to measure the changes of serum hormone levels.Selected bilateral ovary and got rid of surface fat under a microscope in each group.Fixated with ten percent formalin,embedded with paraffin,spliced commonly and HE stained and observed under a microscope.TUNEL assay were used to detect the difference of the apoptosis of ovary granulosa cells in each group. Immunohistochemical analysis was employed to demonstrate the expression of the Bcl-2 and Bax in ovary granulosa cells. Results:①The changes of weigh:the range of weight increasement decreased significantly in treatment group(P<0.001),②serum hormone levels:testosterone levels were significantly lower in treatment group(8.631±4.718ng/ml)than that in control group(16.181±5.314ng/ml)and blank group(7.571±4.326ng/ml)(P<0.001), estradiol levels were obviously lower in treatment group(11.685±6.158pg/ml)than that in control group(15.551±7.632pg/ml)(P<0.05),progesterone levels were obviously lower in treatment group(27.463±10.668ng/ml)than that in control group (33.125±13.147ng/ml)(P<0.05).③histology change of ovary:the number of granular cell layer increaseed obviously up to 4-6 under a microscope in treatment group④apoptosis of ovary granulosa cells:apoptosis rate in treatment group were significantly lower than that in control group(P<0.001);⑤the expression of Bcl-2 and Bax in granulosa cells:the expression of Bcl-2 in treatment group was significantly higher than that in control group(P<0.001),the expression of Bax in treatment group was significantly lower than that in control group(P<0.001).Conclusion:Sodium ferulate can increase the expression of Bcl-2 as well as decrease the expression of Bax obviously in ovary granulosa cells.It can reduce the apoptosis rate of ovary granulose cells remarkably.The number of granular cell layer increaseed and the formation corpus luteum increased.It can diminish the weight of experiment group's animals,reduce the serum testosterone,estradioland and increase progesterone levels obviously.The effects of sodium ferulate on improving endocrine condition of PCO rats and lowing the range of weight increasement may be relevant to its effect of increasing the expression of Bcl-2 as well as decreasing the expression of Bax in ovary granulosa cells,influencing the ratio of Bcl-2 and Bax as well as the formation of heterodimer of Bcl-2,playing the role of antiapoptosis.
