Objective: To investigate the anti-tumor activity of different compounds from three kinds of medicinal plants Selaginella genus, and to elucidate the mechanism.Methods: Experiments were operated in A549, Bel-7402 and HT-29 cell lines inparallel. We used the extracts of Selaginellae genus on human cancer cells.(1)The morphological change of the cells was observed by microscope.(2) MTT assay was used to evaluate the effect of each group compounds on inhibiting cell proliferation. (3)Flow cytometry was used to detect cell apoptosis. (4)Reverse transcript polymerase chain reaction (RT-PCR) was used to observe the level of COX-2 mRNA. Caraphoresis gel photo auto-analysis system was used to detect the products. It was calculated by this formula COX-2mRNA density/6-actinmRNA density.Results: Experiment results showed that some bisflavones of Selaginella labordeihad relatively stronger activities, which depended on dose and time. Flow cytometry further confirmed that inducing cell apoptosis was one of anti-tumor mechanisms. Compare with the negative control group, the total flavonoids groups could significantly inhibit the COX-2mRNA level.Conclusion: Some bisflavones from Medicinal plants Selaginella labordei havestronger action on inhibiting cancer cell proliferation and inducing apoptosis. So it could be concluded that the mechanisms may be that the total flavonoids could inhibit the C0X-2mRNA level .Then could inhibit COX-2 protein expression and inhibit the synthesis of PGE2. Our researches aford some theorical funds for flavanoids from Medicinal plants Selaginella genus to be used in cancer's chemotherapy and will help to broaden the pharmacology values of Selaginella genus.
