| Objective To observe the effects of Glucosidorum Tripterygll Totorum,Tripterygium Hypoglaucum Hutch and Composite Tripterygium Hypoglaucum Hutch on proliferation and apoptosis of human T lymphocyte cell line C8166 and to study the change of cell circle of C8166 cell coculture with CTHH. Methods Cell viability and cell proliferation were measured by typanbhe staining;the inhibition rate and median cytotoxic concentration of C8166 cells were measured by MTT assay;apoptosis was measured by Annexin V-FITC/PI double labelled assay;cell cycle were determined by PI staining and flow cytometry.Results GTT,THH,CTHH could effectively inhibit the proliferation of C8166 cells and the inhibition rate of C8166 cells increased in a time-and concentration -dependent manner;apoptosis rate by flow cytometry increased in a time- and concentration -dependent manner;analysis of cell cycle indicated CTHH mainly leads to the blockage of C8166 cells in G0-G1 stage.Conclusion The cytotoxicity of GTT is larger than THH and CTHH;CTHH inhibits proliferation of T lymphocyte cell C8166 and can induces apoptosis of C8166,leads to the blockage of C8166 Cells in G0-G1 stage in vitro... |
