Objective:To investigate the effects of abnormality expression of Cx43,Cx45 after bone marrow mesenchyma stem cell transplantation in acute myocardial infarction(AMI)in rats.Methods:60 Wistar rats were randomly divided into four groups:normal control(N),sham operation group(SH),injecting of Sodium Chloride after myocardial infarction(MI),injecting of bone marrow mesenchyma stem cells after myocardial infarction(MSCs).The animal model of AMI was established by ligation of the left anterior descending(LAD)coronary artery through thoracotomy.The AMI model was qualified for the elevation of related ST segments of electrocardiogram more than 0.2 mV after operation.Bone marrow was sterilely collected from the bilateral thighbone of rat.Menchymal stem cells were harvested by Percoll method plus adherence method.Third generation of mesenchymal stem cells were injected into the infarcted area and border-zone of the infarcted area after 7days of infarction in rat.The rats were sacrificed after 12 weeks.The heart tissue of nomal area,infarcted area and border-zone of the infarcted area was kept under -80℃.The mRNA expressions of Cx43 and Cx45 was quantified by reverse transcription - fluorescence quantitative -polymerase chain reaction(RT-FQ-PCR).Results:In nomal heart tissue,the mRNA expression of Cx43 and Cx45 among each groups are no difference.Comparing with N group and SH group, the mRNA expression of Cx43 and Cx45 decrease in the infarcted area and border-zone of the infarcted area(P<0.05);In MSCs group,the mRNA expression of Cx43 increases comparing with MI group in border-zone of the infarcted area,but there is no difference in the infarcted area.Comparing with N group and SH group,the mRNA expression of Cx45 in MSCs group and MI group significantly decreases in the infarcted area(P<0.05),but there is no difference between infarcted area,and border-zone of the infarcted area in MSCs group.Conclusion:AMI can decrease mRNA expression of Cx43 and Cx45; Exogenous MSCs transplantation for myocardial infarction can upturn mRNA expression of Cx43 in the border-zone of the infarcted area;but no influence on mRNA expression of Cx45 in both infarcted area and border-zone of the infarcted area.
