| Background and Objective:Nasopharyngeal carcinoma(NPC)is one of the most common malignant tumors in the southen part of China.It is a highly metastatic malignant tumor.As a kind multifunction cytokines,TGF (transforming growth factor)play a key role in cell proliferation,development,transformation and differentiation by integreting with the receptor.The number of transforming growth factor-β1(TGF-β1)has been reported to promote tumorigenesis and increase metastasis directly and/or indirectly in progression, by the absence of the Transforming growth factor-βreceptors(especially the typeⅡ).CD1a is reported as the best marker for imDC.We use CD1 a to study the number and location of imDC in non-keratinizing NPC.The expression of TGF-β1 and TβRⅡin NPC has not been studied well and the relationship of TGF-β1 and DC(dentritic cell,DC)is least understood.In our study,we would like to investigate the expression of TGF-β1,TβRⅡand DC in NPC, and their relationship with pathology and clinic.Materials and Methods:1,Materials.All of the 62 archival histologic specimens of non-keratinizing NPC and 16 adjacent tissue specimens which are choosed from non-keratinizing NPC cases were abtained from the department of pathology of the first hospital affiliated to GuangXi Medical University from 2005 to 2006, 10 cases with chronic nasopharyngitis(CN)as control.2,Methods:①Immunohistochemical methods(ElivisionTM)were used to examine the expression of TGF-β1,TβRⅡand CD1a proteins in paraffin-embedded specimens in 72 specimans,including 62 NPC,10 CN.The results were used to analyse the relationship among the expression levels of TGF-β1,TβRⅡand CD1a proteins,as well as the relationship between the results and the clinicopathologic characteristics of NPC patients.②In situ hybridization(ISH):ISH was used to detect the expression of CD1a mRNA in specimans above.The relationship between the result and the clinicopathological fatures were analyzed.Results:1,Immunohistochemistry reveal that expression rates of TGF-β1 protein were 67.7%(42/62)in NPC,93.8%(15/16)in paratumorous tissues,and 50% (5/10)in the control group.The positive rate of TGF-β1 protein in NPC was significantly correlated with the rate in control group and paratumorous tissues (P<0.05).The expression of TGF-β1 protein in NPC was significantly correlated with the TNM staging and the lymphaden metastasis(P=0.033,P= 0.008),but not correlated with the other clinicopathological features(P>0.05).2,Immunohistochemistry reveal that expression rates of TβRⅡprotein were 58.1%(36/62)in NPC,100%(16/16)in paratumorous tissues,and 70% (7/10)in the control group.The positive rate of TβRⅡprotein in NPC was significantly correlated with the rate in paratumorous tissues(P<0.05).The expression of TβRⅡprotein in NPC was not correlated with all the clinicopathological features(P>0.05)3,Immunohistochemistry reveal that expression rates of CD1a protein were 41.9%(26/62)in NPC,12.5%(2/16)in paratumorous tissues,and 0%(0/10)in the control group,respectively(P<0.05).The expression of CD1a protein in NPC was significantly correlated with the lymphaden metastasis(P=0.011),but not correlated with the other clinicopathological features(P>0.05) 4,ISH reveal that expression rates of CD1a mRNA were 33.9%(21/62)in NPC,6.25%(1/16)in paratumorous tissues,and 0%(0/16)in the control group. The expression of CD1a mRNA in NPC was significantly correlated with the TNM staging and the lymphaden metastasis(P<0.05),but not correlated with the other clinicopathological features(P>0.05)5,By Spearman correlation analysis,the expression of CD1a and TGF-β1 protein is positively correlation(r=-0.304,P=0.016);the expression of TβRⅡand TGF-β1 protein is positively correlation(r=-0.272,P=0.033);the expression of CD1a protein and CD1a mRNA is positively correlation(r= 0.533,P<0.0005)too.Conclusions:1,TGF-β1 promotes tumorigenesis and increases metastasis in non-keratinizing carcinoma of nasopharynx tissue.This result suggests that the deletion of TβRⅡmay be one of the reasons of cells'escaping the negative regulation of TGF-β1.2,The number and morphous of imDC marked by CD1a can reflect the partial condition of patien's immune system.The expression of CD1a may predict metastasis potential and the development of non-keratinizing carcinoma of nasopharynx.3,TGF-β1 can reduce the number of imDC and inhibit the imDC's function.They might promote the development of the non-keratinizing carcinoma of nasopharynx together. |
