| Objective To investigate the lymph node metastasis(LNM) -associated protein in endometrial carcinoma(EC).Methods Tissues from 4 normal endometria(Groupâ… ), 4 non-LNM EC(Groupâ…¡)and 4 LNM EC(Groupâ…¢) were analyzed by two-dimensional gel electrophoresis. The differently expressed proteins were identified by mass spectrometry (MALDI-TOF-MS).Results Eleven proteins were found to be differentially expressed between non-LNM EC and LNM EC. Each of them was identified. Among them, the heat shock protein 27, actin, S100 calcium-binding protein A9, pyruvate kinase, alpha enolase, serum albumin, histidine triad nucleotide binding protein 1 were up-regulated, while the ASRGL 1 protein and Esterase D/formylglutathione hydrolase drow-regulated compared with that in the non-LNM EC.Conclusions Significant difference of proteins express was found in the non-LNM EC and LNM EC by two-dimensional gel electrophoresis. Nine proteins identified may play a role in the process of lymph node metastasis of EC. The differently expressed protein between endometrial carcinoma with and without lymph node metastasis could be identified by two-dimensional gel electrophoresis coupled with mass spectrometry. |
