The Expression And Significance Of LRP16 Gene And In Human Meningioma

Objective:①To detect the quantity of both LRP16 mRNA and LRP16 protein in meningiomas resected off the patients with the tumor.②To analyze the immunoreactivities of sex hormone receptors in meningiomas,as well as PCNA and FVⅢ-Rag,and the correlations among them.③To explore the functions of LRP16 gene in meningioma,by correlating LRP16 mRNA with sex hormone receptors,as well as PCNA-LI and MVD.Methods:①The resected tissues,of both and dural mater,and the clinical data of patients were collected,and then the mRNA level of LRP16 in the tissues was detected by using both RT-PCR and real-time PCR method.②Then,the quantity of LRP16 protein was the detected by Western Blot method in the same samples.③Subsequently,we used immunohistochemical method to evaluate the immunoreactivities of sex hormone receptors in meningiomas,as well as PCNA and FVⅢ-Rag in the same samples.④Finally the relevant statistical methods were introduced to analyze the relations among the indexes involved above.Results:①The results of conventional PCR showed that 71.4%(30/42)samples in the tumor group present positive,compared with 16.7%(7/42)weak positive and 11.9%(5/42)negative.While all the cases of the in the control group appeared negative.②The results of real-time PCR was found that the relative quantification(LRP16/GAPDH)of mRNA expression for LRP16 gene in the tumor group was about 0.205±0.134,higher than 0.003±0.001 of the control group with statistical significance(P＜0.01).③According to the results of Western Blot,the OD ratios(LRP16/β-actin)of bands for cases in the tumor group was higher than that in the control group(P＜0.01).④As to the immunohistochemical results,69.0%(29/42)of the menigiomas related showed PR positive,while none of which was detected AR or ER positive.Positive rate of PR in the benign tumor(WHOI)group was higher than that in the non-benign(WHOⅡandⅢ)group(P＜0.05).While PCNA-LI and MVD of tumors in the non-benign group were higher than that in the in the benign group but no differences was found between PR(-)and PR(+)group.⑤The relative quantification of LRP16 mRNA in the non-benign group was higher than the benign group(P＜0.05),but no differences between PR(-)and PR(+)group.And the mRNA expression of LRP16 gene was correlated with PCNA-LI(P＜0.05, r=0.8329)but MVD.Conclusion:①LRP16 gene is high expressed in menigiomas and may contribute to behaviours of tne tumor.②The expression of LRP16 is related to menigioma's grades of malignancy.③The expression of LRP16 is correlated with cell proliferation of menigioma and LRP16 may contribute to the regulation of cell proliferating.
