The Effect Of Donor's Immatura Dendritic Cells To Rat Skin Allograft

ObjectiveTo build the model of allograft rejection after rat skin transplantation. And found a simple method to expand the immature dendritic cells. Then we studied the effect of donor's immature dendritic cells to rat skin allograft.MethodFirst, we practiced the skills of rat skin transplantation with rat skin self-transplantation. And we established a new simple method for increase the number of immature dendritic cells for the study: First, we incentive the imDCs in vivo by used rhG-CSF as 5μg/kg 7 days. Second, the imDCs from rat bone marrow were cultured 3 days in vitro. Third, reject them into the receptor from vena dorsalis penis at the same time of rat skin allograft. We observed the survival time and the pathology of skin allografts according to the groups. And we discussed the effect of The effect of donor's immatura dendritic cells to rat skin's allgraft.Result1. All the skin of self-transplantation were had a good condition (>2month).The allograft of rat skin were matched with the skin of donor's. There was no loss. And there was is little difference. The subsidiary hairs and hair follicles in allograft were healthy. The pathological section of allograft after 14 days showed that each organization layer clear and integrity, and the subsidiary structure was intact. The structure of organize cell was integrity. And there was few immunity cells. 2. Control-l(Wistar→SD):all the allografts were rejected in 14 days. The survival time was 11.8±1.6 days, and the least was 9 days, the longest was 14 days. 3.Control-2(Wistar→Wistar) group: Though the skins were rejected at last, they have a long survival time, and the graft rejective reaction was light. The survival time is 23.1±2.4 days,19 days to 27 days. The pathological section of allograft after 14 days showed that the structure of organization structure has been destroyed. We could found the necrotic tissue, and large number of immunity cells. We could found the caryolysis and nuclear fragmentation at high enlarge times.4.The imDC group: the allograft of skin' survival time is 19.3±3.4 days, 14 days to 25 days. The pathological section of allograft after 14 days showed that the structure of organization structure has been destroyed. We could found the necrotic tissue, and large number of immunity cells. We could found the caryolysis and nuclear fragmentation at high enlarge times. And, we had a conclusion that the group of imDC had a more ligher graft rejective reaction than control-1 group from pathological sectionhe.ConclusionThe pretreatment with imDC can prolong the survival time of rat graft after transplantation, and lighten the graft rejective reaction.
