With the development of biomedical engineering, the biological medical material attracts people's attention increasingly. As a kind of natural biomaterial, collagen has low antigenicity, biodegradability, good biocompatibility and can promote cell migration and proliferation. So it is widely applied in the biological medical material domain. Human-like collagen (HLC) is one kind of new protein high-density fermented by the genetic engineering bacterial and extracted. Preparing hematischesis material with HLC to solve the problems as follows: the hematischesis effect is not ideal, to degrade slowly, the tissue repair function is not obvious and so on, that is our conception. Using glutaral to crosslink HLC to get sponginess hematischesis material, and it is named Human-like collagen hematischesis sponge. The physical and chemical properties, hemostatic effect and biocompatibility of human-like collagenhematischesis sponge was studied.1. Using HLC as the fundamental ingredient to cross link with glutaral and usingvacuum freeze-drying method to get hematischesis sponge. Through the experiment we could get the result as follows: the sponge internal aperture probably is 100~300μrn, the porosity is above 90%, the water-absorptivity approximately is 3100%, the density is 0.043 g/cm~3, the ash is less than 0.1%, the shape is complete in the physiological saline with 8 weeks, the microorganism has not picked out, the endotoxin test result is the negative, the sponge degrade completely in vitro with 0.05% type I collagenase, 37℃, within 2h, the physical and chemical properties is very good;2. The cytological experiments indicated that 2BS cell could grow and multiply normally on the surface of HLC hematischesis sponge, the crepping action and the cell morphous was normal, which showed that Cell compatibility is good; 3. In the animal model, the hol-hemostatic time of human-like collagen hematischesis sponge to the rabbit ear trauma was 79±8s, to the rabbit liver trauma was 51±7s, the hematischesis effect has the significant difference compared with the similarly hematischesis material applied in the clinics (P<0.05);4. Human-like collagen hematischesis sponge, DOUBLE DIAMOND collagen sponge and absorbable gelatin sponge were planted in the rabbit liver, 2 weeks, 4 weeks and 8 weeks later, the hepatic tissue was analyzed by HE staining and transmission electron microscopy, the result indicated that Human-like collagen hematischesis sponge degrade more quickly, and has less effect to the structure and function of hepatocyte, hepatic tissue, hepatic tissue has no pathological changes, hepatic histocompatibility of human-like collagen hematischesis sponge was superior to DOUBLE DIAMOND collagen sponge and absorbable gelatin sponge.
