Objective:The growth and metastasis of tumor have vessel dependence. New vessels are important factors in tumor growth, invasion and metastasis. Of all, VEGF and angiostatin(AS) play important roles in angiopoiesis. Now, most researches in and abroad focus on how AS influence the tumor new vessels and the exploratory development of AS in tumor anti-angiopoiesis therapy. But there are few researches in the relationship between AS and tumor's formation, development, metastasis, prognosis. The study is to identify the effect and clinicopathologic relationship of AS'and VEGF's mRNA in NSCLC carcinogenesis and development by detecting the mRNA expressions of AS and VEGF.Empirical method:A total of 48 lung cancer tissues and paratumor lung tissues(the distance to tumor above 5cm) obtained from the surgical resection at the pathological department of the forth Affiliated Hospital of China Medical University between the September 2006 to June 2007, liquid nitrogen freezing, storage at -80℃. In 48 cases of lung cancer, the ratio of male to female is 33:15, the ages are between 41 and 85, the average age is 62. According to the World Health Organization lung and pleural tumors classification criteria(1999), squamous carcinoma is 17 cases, adencarcinoma is 22 cases and others are 9 cases. TNM(the stage standard is UICC1997): I stage: 12 cases, II stage: 18 cases, III stage: 16 cases,Ⅳstage: 2 cases. All cases have integrated clinical and pathological data. We used RT-PCR technology , 2% garose gel electrophoresis, dyeing by ethidium bromide, and observe the result by viltalight lamp. Judging the fragment by DNA Marker, imaging analysis the amplification product, and measuring the OD value, we can quantitively analyze throughβ- actin. The results were analyzed by SPSS 12.0 package, the measurement data was signed with ?x±s , using t-test and pearson correlation analysis. P<0.05 was considered statistically significant. P<0.01 was considered striking significant.Results:1. mRNA expression of AS in NSCLC and paratumor lung tissues:The mRNA expression of AS in NSCLC is 7.86±1.73 and the mRNA expression of AS in paratumor lung tissues is 5.56±1.40. The mRNA expression of AS is higher than that in paratumor lung tissues( P<0.05).2. Relationship between mRNA expression of AS and NSCLC clinicalpathological characteristics:The mRNA expression of AS in I-II stage lung cancer is higher than that in III-Ⅳstage lung cancer; The mRNA expression of no metastases is higher than that of metastases, the difference has statistical significance(P<0.05). There was no significant correlation between the mRNA expression of AS and patients'age, gender, pathotyping and grading(P>0.05).3. mRNA expression of VEGF in NSCLC and paratumor lung tissues:The mRNA expression of VEGF in NSCLC is 12.43±1.58, The mRNA expression of VEGF in paratumor lung tissues is 9.10±1. 20. The mRNA expression of AS is higher than that in paratumor lung tissues( P<0.01).4. Relationship between mRNA expression of VEGF and NSCLC clinicalpathological characteristics:The mRNA expression of AS in III-Ⅳstage lung cancer is higher than that in I-II stage lung cancer( P<0.01); The mRNA expression ofⅢgrading is higher than that ofⅠ—Ⅱgrading ( P<0.05); The mRNA expression of metastases is higher than that of no metastases, the difference has statistical significance( P<0.01). There was no significant correlation between the mRNA expression of VEGF and patients'age, gender and pathotyping(P>0.05).5. Correlation between AS and VEGF expression in NSCLC:The result of correlation test between AS and VEGF: r=-0.161,P=0.274 (P>0.05).They have no correlation.Discussion:The invasion and metastasis of tumor have close relationship with the formation of new vessels and the degradation of stromous. Tumor vascularization depends on the balance of promotive angiogenesis factor and suppressive angiogenesis factor. Human AS can inhibit endothelial cells proliferation and metastasis. It can also enter blood circulation and effect on locus far from tumor. VEGF family is a kind of multi-functional cytokine and reacts in many diseases. It has been regarded as an important index in measuring tumor's growing and metastasis. The results show that expressions of AS and VEGF are both higher than those in paratumor lung tissues. The difference has striking significance. Different from VEGF, the expression of AS in NSCLC decreases with the increase of clinical stages. Expressions with metastases are lower than those without metastases. It demonstrates when NSCLC's stage is later, the metastasis rate is higher and the expression of AS is lower. The study also shows AS has no correlation with mRNA of VEGF. The mRNA expression of VEGF has no relationship between their expressions and the patient's age, gender, and the tumor's pathotyping, but the mRNA expression of poorly differentiated is higher than that of well-moderately differentiated, these is concord with literature's report.Conclusion1. The mRNA expressions of AS and VEGF in NSCLC are higher than those in paratumor lung tissues, and there was correlation between the level of AS, VEGF and lymph node status and pTNM.2. The expressions of AS in NSCLC depressed gradually with increasing of stage of pTNM. The expressions of AS in lymph node diversion group were lower than that of AS in non lymph node group. The expression of AS mRNA was opposite with that of VEGF mRNA, and these are the result of their interaction.3. The experiment hints that the expression of AS mRNA and VEGF mRNA in NSCLC tissue can reflect the capability of invasion and transfusion in some degree. They may correlate with development and advancement of NSCLC.
