Objectives:To observe the effect of neural stem cell on the expression of basic fibroblast growth factor(bFGF) after acute spinal cord injury and its significance.Methods:70 SD rats were divided into the groups at random as follow:(1)normal control group : 10 rats . only expose the putamen.(2) spinal cord injury control group(2d group): 10 rats ; (3)spinal cord injury control group(5d group) : 10 rats ; (4) spinal cord injury control group(12d group) : 10 rats ;were divided into three group . (5)spinal cord injury stem cell group(2d group) : 10 rats ; (6)spinal cord injury stem cell group(5d group) : 10 rats ; (7)spinal cord injury stem cell group(12d group) : 10 rats. Taking the materials : About 8 mm ispinal cord that the central position of it is the injuried area was taken for HE staining and immunohistochemical staining and Nissl's staining respectively. Observe the behavior and histological and immunohistochemical changes . Observe the dynamic changes of bFGF in the above mentioned groups with immunohistochemical staining , and staining was quantitated by imagine analysis technique.Results:1. In behavior examination , there was significant difference (P<0.05) between the spinal cord injury stem cell group and the spinal cord injury control group . 2. Histological examination : In the spinal cord injury control group , 2d after spinal cord injury , there were splinter haemorrhage in the gray mater and white mater , swelling of axon , fragmentation of myelin sheath and the damage of nerve cell ; 5d after spinal cord injury , there were proliferation of glia cell , endotheliocyte and lymphocyte in the zone of injury ; 12d after spinal cord injury , the proliferation is obvious . In the spinal cord injury stem cell group , 2d after spinal cord injury , there were small splinter haemorrhage in the gray mater and white mater , the degree of swelling of axon , fragmentation of myelin sheath and the damage of nerve cell was lower than the spinal cord injury control group ; 5d after spinal cord injury , there were more glia cell in the zone of injury than the spinal cord injury control group ; 12d after spinal cord injury , there were more glia cell in the zone of injury than the spinal cord injury control group.3. Expression of bFGF:In 2d , 5d and 12d after spinal cord injury , the expression of bFGF and the number of bFGF positive cell were progressively increased . There was significant difference (P<0.05) between the spinal cord injury group and the normal control group . There was significant difference (P<0.05) between the spinal cord injury stem cell group and normal control group . There was significant difference (P<0.05) between the spinal cord injury stem cell group and the spinal cord injury control group.Conclusion: 1. 2d , 5d and 12d after spinal cord injury , the expression of bFGF and the number of bFGF positive cell were progressively increased . Application of the neural stem cell can induce the increasing of bFGF expression. It not only acted on the receptor of neuron and glia cell, made the axon of neuron regeneration and the glia cell proliferation ; but also bFGF can proliferate the neural stem cell , because the neural stem cell can differentiate into neuron , glia cell and glia precursor , all these were favourable to the repair of spinal cord injury . 2. Application of the neural stem cell can lower the degree of axon swelling , fragmentation of myelin sheath and the damage of nerve cell, and then it lowered the degree of spinal cord injury , and made the glia cells proliferation in the injury zone , so it promote the repair of spinal cord . 3. Application of the neural stem cell can promote the recovery of the feeling and motor function of the rats with spinal cord injury.
