Objective:Diabetic nephropathy is a common complication of diabetic microvascul -ar complications and is the most important single factor in chronic renal function failure .Patient of hemodialysis and renal transplantion every year .one-forth patients are caused by diabetic nephropathy. Diabetic nephropathy mainly therapy is control meat and drink ,control blood pressure ,drop lipoids,reduce proteuria .But there are still many eager making advancement to end stage renal failure. With the cytobiolog -y and molecular biology development, a lot of reseaches have discoved that infla- mmatory cell infiltrate, many cell factors and mediators of inflammatory increasing have effect during the development of diabetic nephropathy.Glomerular fibrosis of diabetic nephropathy is glomerular extracellular matrix abnormal assemble.Researches show transforming growth factor-β1 (TGF-β1) and connective tissue growth factor (CTGF) are important growth factor in glomerular fibrosis progress. Cordceps sinensis (CS) improve cell metabolize ,regulate fatty metabolize , anti -oxidize,anti-fibrosis in liver, whether CS has anti-fibrosis function,Nowdays resear -ches are scarce .This experimentation uses streptozotocin (STZ) to induce diabetic nephropathy modle in rat. Making CS to cure diabetic nephropathy . We also observ - ed the effects of CS on the expression of TGF-β1 and CTGF in the renal tissue of DN rats, and to investigate the anti-fibrosis mechanism of CS.Methodology: Thrity-six male Wistar rats were randomly divided into normal control group,DN group and treated group with CS[5.0g/(kgd)]. At week2, week4 and week8,four rats of three groups were sacrificed to test renal fibrotic injury, serum creatinine(SCr) and 24h urinary protein count. The expression of TGF-β1 and CTGF were examined by immunohistochemical technique. Result::Compared with N modle group, the levels of SCr, 24h urinary protein excretion was raised significantly in DN modle group ( P<0.01). compared with N modle group, the levels of SCr, 24h urinary protein excretion was raised significantly in CS treatment group ( P<0.01).compared with DN modle group, the levels of SCr, 24h urinary protein excretion, and glomerular fibrosis was reduced significantly in CS treatment group ( P<0.01). At the same time, the expression of TGF-β1 and CTGF was significantly decreased in CS treated group (P<0.05 or P<0.01).Conclusion:CS can reduce inflammatory cell infiltration cell infiltration in experimental diabetic rats and decrease serum creatinine,24h urinary protein of diabetic rats . CS also can downregulating the expression of glomerular and tubular TGF-β1 and CTGF expression.
