| Objective:To investigate TNF-αexpression in non-small cell lung cancer of different clinic stages, and the association of TNF-αpolymorphism with susceptibility to the severity of non-small cell lung cancer.Methods:1.ELISA method was used to dectect TNF-αlevel in serum.2. PCR-RFLP method was used to analysis TNF-αpolymorphism.Results:1.Compared to control group,TNF-αlevels were significantly different in the serum of non-small cell lung cancer patients. Also, TNF-αlevels were different in different stages. But, there were no difference between AD group and SC group. 2. There were significant differences in TNF-α-308G/A, TNF-α-238G/A genotype distributions between controls and uung cancer patients. But, there was no association between the distributions of genotype and clinic stages or pathological type.Conclusion:1.Compare to control group, TNF-αlevels were significantly higher in serum of non-small cell lung cancer patients. As the disease got worse, the levels of TNF-αgot higher and higher. Accordingly, detecting the TNF-αserum level can help to judge immune state and predict the disease. 2. A association between the TNF-α-308G/A and TNF-α-238G/A polymorphisms and the susceptibility to non-small cell lung cancer was demonstrated. The -308A allele has a promotive effect for non-small cell lung cancer development, whereas the -238A allele has a protective function against non-small cell lung cancer. |
