Objective1 To observed the change of the myocardium enzyme,malonaldehyde(MDA),nitric oxide synthase(NOS),which happened in the process of the immature myocardial ischemia reperfusion injury.2 By the change of the myocardium enzyme,MDA,and NOS,we could observe if the xinmailong have had the protection for immature rabbit myocardial ischemia reperfusion,and explore the mechanism.Method1 Grouping and model building:54 immature rabbits were complete randomly divided into three treated group:Sham group,ischemia reperfusion(I/R) group and xinmailong(XML) group.Each treated group consisted of 18 rabbits. Evenly treated group randomly divided into three time point,each time point has 6 rabbits.The experimental model of anesthetized immature rabbit MIRI was established by ligated the left diagonal branch of coronary artery for 30min followed by reperfusion for 60min,120min and 240min.2 To observed the effect of xinmailong on the amount of MDA.NOS.myocardial enzyme and the myocardial fine structural.Results1 myocardial enzyme①Creatine kinase(CK):in I/R group and XML group, the CK were all evidently higher than Sham group(p<0.01).At every reperfusion time point, the increased argument of CK in XML group was lower than I/R group(p<0.01).②The rest myocardial enzyme(AST.LDH.HBDH):With the extend of reperfusion time,the rest myocardial enzyme were all increased in I/R group,which higher than Sham group(p<0.05或p<0.01),were all was slightly increased in XMLgroup,which compared with Sham group,had no significant deviation(p>0.05),and evidently less than I/Rgroup(p<0.01).2 MDA Compared with sham group,the content of MDA in I/R group and XML group were all evidently increased(p<0.01).The content of MDA had reached the peak at 1 hour after reperfusion,which consisted beyond 3 hours in I/R group and XML group.Compared with I/R group,the XML could evidently decrease the content of MDA(p<0.01).3 The activity of NOS①The activity of ecNOS in XML group and I/R group were all evidently lower than the Sham group(p<0.01),but the decreased argument of the activity of ecNOS in XML group significient lower than the I/R group(p<0.01).②1 hour after reperfusion,the activity of iNOS in I/R group and XML group were slightly increase,which compared with the Sham group had no significant deviation(p>0.05).The activity of iNOS in I/R group and XML group were all higher and higher with the time extending after reperfusion.But the activity of iNOS was no significant deviation between the two group(p>0.05).4 The myocardial fine structural After ischemia reperfusion,mounts of inflammatory cell infiltrated,myocardial cell degeneration and necrosis.XML could inhibit the infiltration of inflammatory cell,lighten the myocardial cell degeneration and necrosis.Conclusion1 After ischemia reperfusion,the the myocardial enzyme(CK.LDH.HBDH.AST),the content of MDA and the activity of iNOS were all increased,the activity of ecNOS decreased.2 XINMAILONG has protective effects on MIRI.The protective effects maybe that inhibit lipid peroxidation,and decrease the production of oxygen free radical,and protect the endothelial of coronary artery,and increase the content of nitric oxide,and lighten the structure injury and dysfunction.
