ObjectivesTo investigate the expression of vascular endothelial cell adhesion molecule-1 (VCAM-1) on human umbilical vein endothelial cell line (ECV-304) stimulated by serum of preeclampsia and interfered by basic fibroblast growth factor (bFGF), determine whether bFGF have protective effect on vascular endothelial cell damage and provid a new mathod to treat hypertensive disorders in pregnancyMethodsECV-304 were cultured in culture flasks in DMEM with 10% fetal calf serum(FCS),then change following medium after ECV-304 reached confluence.(l) nomal blood serum medium(NP group) (2) preeclamptic blood serum medium(PE group) .(3) blood serum medium containing preeclamptic serum and bFGF with concentrati- ons 5,10,50ng/ml(PE+bFGF group).(4)negative control (containing 20% fetal calf serum, proteic concentration is 5 ng/ml,PH7.2).after culturing 24 hours ,detect the content of VCAM-1 on vascular endothelial cell with Immunocytochemical staining and Reverse transcription polymerase reaction (PCR)Result(1) ECV-304 express more VCAM-1 in PE group compared to NP group (P<0.05).(2) ECV-304 in PE+bFGF group express less VCAM-1 than that in PE group(P<0.05) and have dose-dependentConculusionECV-304 can express more VCAM-1 when they were stimulated by the serum of preeclamptic patient ,However, when they were cutured with bFGF ,the level of VCAM-1 was repressed. It suggest that extrinsic source bFGF may have protective effect on vascular endothelial cell damage in preeclampsia,and it may be valuble for treating preeclampsia. |
