Objective:To investigate the expression of signal transducers and activators of transcription 3(STAT3)in human oral squamous cell carcinoma and to evaluate the relationship between protein level and clinicopathological parameters and the probability of the STAT3 using in the gene therapy. Methods:The immunohistochemistry SABC was used to detect the expression of the stat3 in human oral squamous cell carcinoma and precancerous lesion. The automatic image analyser was used to analyse the gray scale of the STAT3.Result:STAT3 protein level was significantly increased in squamous cell carcinoma tissue as compared with precancerous lesion(P<0.05).There were statistical differences of the average gray scale between the different differentiated cancers of the STAT3 protein(P<0.01).The expression of STAT3 was higher in poor-differentiated cancers than in well-differentiated cancers. The expression of STAT3 was significantly different between the T1,T2 and T3(P<0.01).Metastatic lymphnodes overexpressed STAT3 protein as compared with non-metastatic lymphnodes.Protein level of STAT3 in the stageⅢand stageⅣcarcinomas was higher than that in stageⅠand stageⅡcarcinomas(P<0.01).Conclusion:Overexpression of STAT3 may play an important role in the carcinogenesis, development and the clinical stage of the human oral squamous cell carcinoma and it may be one of the indexes of the diagnosis and therapy.
