Spermatogenesis is a complex process of cell division and differentiation resulting in a series of molecular and morphological changes in male germ cells. This process requires the highly regulated expression of a number of genes. Zinc finger family is one of the biggest family in the mammalian genome, which plays an important role in spermatogenesis and spermiotelcosis.We investigated the function of spermatogenesis-related gene-ZNF230 in the present research, including two parts below.To construct a conditional targeting vector of mouse zinc finger protein gene Znf230. Specific primers were designed and used to amplify two homologous arms of 4 kb and targeting sequence of 1kb from mouse genome. Then cloned amplicans into pBS vector with Neo gene in reverse direction. After construct analysis by REase and sequencing, the coincidence of fragment in vector with the sequence in Genebank was confirmed (NMJ325999). For constructing conditional targeting vector by which PCR and directional cloning is a simple and reliable method. We obtained the construction of mouse zinc finger protein gene Znf230 conditional targeting vector.Cancer-testis (CT) antigens are presently considered as targets for the development of cancer immunotherapy protocols. CT antigen genes are specifically expressed in tumors, but not in normal tissues except the testis. ZNF230 is a zinc-finger protein gene located on the 11p15 which was first identified and cloned from human adult testis. In the second part of the report, the expressions of ZNF230 mRNA and protein were examined in tumors with different histological types. It was found that ZNF230 mRNA was expressed in a variety of tumors including those of lung, liver, kidney, esophagus, stomach cancers and so on, indicating that ZNF230 is a novel member of CT antigen family.
