| Objective To investigate the change of hypoxia on the expression of hypoxia inducible factor 1αmRNA and protein in pancreatic cancer cell PANC-1 cell line.Methods The hypoxia culture model was established by a hypoxic chamber.The gene productions of HIF-1 were observed at different hypoxic culture phase.PANC-1 cells were divided into four groups: control group;hypoxic group(24h);hypoxic group(48h);hypoxic group(72h).The cells were harvested at different time,The mRNA expression of HIF-1 was showed by the semi-quantitative reverse transcription PCR(RT-PCR). The HIF-1a protein was tested using western blot analysis.Results After the hypoxia treatment,comparing with control group,the mRNA and protein expression of HIF-1a increased significantly(P<0.01) in hypoxic groups. The protein and mRNA expression was increased remarkably ( P < 0.01 ) with respondence to the hypoxia time. Conclusion The environment hypoxia can induce the expression of HIF-1αin pancreatic cancer PANC-1 cell line . The mRNA and protein expression of HIF-1a increased significantly with respondence to the hypoxia time. |
