Object To establish rat coronary microthrombosis medel; To investigate the possible mechanism of microthrombosis and elucidate the effection of microthrombosis on heart function.Methods The rat model of coronary microthrombosis is developed by injecting sodium laurate into ascending aorta during clamping aorta. In the sham operation group, equal volume 0.9% sodium chloride was injected instead of sodium laurate. Myocardial sections stained with hemotoxylin and eosin was observed to determine whether there are microthrombosis in rat coronary arterioles. The plasma contents of vWF was also measured. A serial echocardiography was performed to monitor the alterations of cardiac function. Expression of fgl2 in myocardial tissue was measured by RT-PCR.Results 1 Microscopic examination of myocardial sections revealed platelet-fibrin microthrombi in experimental group while no changes in sham operation group. 2 The plasma contents of vWF was beginning to increase one hour after operation and return to its normal level after one month while no changes in sham operation guoup. fgl2 was beginning to increase after one hour and got its highest level after one week, while no expression in normal or sham operation group. 3 Comparing with sham operation group, LVIDd and LVIDs were significantly enlarged after one month; EF and FS were significantly decreased after one week. The level of EF and FS were highly correlated with the level of fgl2.Conclusion 1.Sodium laurate can successfully induce rat coronary microthrombosis which provide a economy animal model to investigate no-reflow phenomenon. 2 .Sodium laurate can injure endothelial cells, and fgl2 may participate the process of microthrobosis. It is may one of the mechanisms which endothelial cells promote coagulation in pathologic conditions. 3. Myocardial microthrombosis can induce EF and FS decrease and heart chambers enlargement.
