Objective:To study the mechanisms of gossypol acetate inhibiting actions andinducing apoptosis on RM-1 prostate cancer cells. Methods:MTT assay was usedto identify the effect of gossypol acetate on RM-1 cells in vitro. RM-1 cellapoptosis was detected by electron micrographs, Hoechst33258 and flowcytometry. Bax protein and mRNA were observed by FCM and RT-PCRassay. ResultS:The IC50 value of gossypol acetate for RM-1 were found to be30.56mg/L.The apoptotic cells were induced by gossypol acetate at the concentrationof 30mg/L.RT-PCR assay indicated that the percentage of apoptotic cells and thepositive cells of Bax protein and mRNA were increased with the increasing ofexposure time of gossypol acetate. Conclusion:gossypol acetate can inhibit RM-1prostate cancer cells and induce apoptosis at some concentration.The importantmechanism were enhancing the level of bax protein and up-regulating mRNA of baxgene.
