Objective: To investigate the relationship between Genetic polymophisms ofConnective tissue growth factor promoter and HBV-hepatic cirrhosisMethods: Genetic polymorphisms of connective tissue growth factorpromoter (locus-484G/C and-650G/C) were determined in 121 patients withHBV-hepatic cirrhosis and 138 healthy blood donors by polymerase chainreaction (PCR) and direct capillary sequencing.Results: The frequency of polymorphisms of CTGF-promoter-484T/C wasnone in all people. There was no significant difference in genotypedistribution at CTGF-promoter-484T/T between the two groups. Thefrequency of CTGF-promoter-650GC was higher in the HBV-hepaticcirrhosis patients than in the healthy blood donors (10.74%∨4.35%,P=0.049, P<0.05). While among the patients with liver cirrhosis, thefrequency of CTGF-promoter-650GC was higher in the patients withabnormal ALT than with normal ALT (16.42%∨3.70%, P=0.025, P<0.05),and the frequency of CTGF-promoter-650GC was highest in Child-Pugh B group than Child-Pugh A group and Child-Pugh C group(3.57%∨30.43%∨9.52%, P=0.002, P<0.05).Conclusion: Genetic polymorphism of connective tissue growth factorpromoter-650G/C is correlated with HBV-hepatic cirrhosis. The CTGFpromoter-650GC mutation may be the adverse host factor in HBV-hepaticcirrhosis, and play an essential role in the development and progression ofhepatic fibrosis. These findings may have important therapeutic implications.
