Effects of Fosinopril on Transforming Growth Factor-Beta 1 and Matrix Metalloprotein-ases-2Secretion of Glomerular Mesangial CellObjectiveGlomerular mesangium is composed of glomerular mesangial cell (GMC) and extracellular matrix (ECM). The pathologic change of glomerular mesangial is the basis which leads to glomerular sclerosis and renal failure. GMC proliferation (during varies of immune and non-immune injuries) lead to aggradation of big molecular substance on glomerular mesangium and the mechanical obstruction on the glomerular capillaries which reduce the glomerular filtration rate (GFR). How to control the GMC proliferation has great significance in reversion of glomerular injury and chronic change of glomerular disease.ACEI protective effect of kidney is hotspot of research, which plays an important role in protecting the kidney. The experiment reveals the excessive aggradation of ECM is the positive factor of glomerular sclerosis. A great many of researches reflect varies of factors (both the promoting and inhibiting factors) affect the GMC proliferation.To observe the effects of fosinopril(FOS), a new generation angiotensin-converting enzyme inhibitor(ACEI), on protein expression of transforming growth factor-β1 (TGF-β1) and matrix metalloproteinases-2 (MMP-2) of rats glomerular mesangial celI(GMC) induced by lipopolysaccharide(LPS); To demonstrate the effects of GMC multiplication and matrix degradation by applying ACEI; To explore the Effective dose of FOS on Glomerular disease treatment, so as to against glomerular sclerosis. MethodsThe cultured GMC in classic way were divided into 3 groups: control group; LPS group; LPS+FOS group(the concentration of f osinopril was 10—8mol/L,10—6mol/L,10-4mol/L).TGF-β1 and MMP-2 expression in GMC was detected by SABC(Strept Actividin-Biotin Complex); The results were analyzed by Pearson correlation analysis.Results(1) The Positive expression of TGF-β1 and MMP-2 secretion were brown particles localed in cytoplasm. (2) LPS group was obviously higher than control groups in TGF-β1 and MMP-2 secretion expression.. (3) In LPS+FOS group, with the incerasing of the concentration of fosinopril, the expression of TGF-β1 secretion decreased and MMP-2 secretion expression increased distinctively compared.Conclusions(1) FOS has obviously inhibited on TGF-β1 expression of rat GMC and promoted the expression of MMP-2, which reveals that it may be an important mechanism by FOS on restraining the GMC multiplication. (2) GMC multiplication restraining effect of FOS has positive correlation to its concentration.
