Research On The Effect Of The Suanzaoren Decoction-Contained Serum On The Protective Effect Of PC12 Cell Apoptosis Induced By Corticosterone To Simulate The Nerve Cell Impaired Associated With The Anxiety

Objective: To investigate the effect of the Suanzaoren decoction(SZRT)-contained serum on the protective effect of PC12 cell apoptosis induced by Corticosterone(CORT) and the related regulatory protein, and probe into the molecular mechanism of anti-anxiety effect of SZRT, to lay the foundation of the research and development of anti-anxiety Chinese medicine.Methods: To simulate the nerve cell impaired associated with the anxiety with the PC12 cell apoptosis induced by high level CORT. To observe the PC12 cell proliferation under condition of CORT and SZRT-contained serum through MTT method; to observe the effect of SZRT-contained serum on PC12 apoptosis through flow cytometry and Giemsa staining; to determine the Caspase-3,Bcl-2,NF-кB expression with immunocytochemical method.Results: SZRT-contained serum can make the PC12 cell proliferation rate increased and apoptotic rate decreased significantly. The Giemsa staining shows that the whole cell shapes and apoptotic bodies were observed occasionally. The results of immunocytochemical method show the Caspase-3 expression reduced significantly, Bcl-2 expression increased obviously, but there was no significant difference in the Bcl-2 expression compared with the model. The results of NF-кB immunocytochemical staining had no difference among the all experimental groups. Conclusion: SZRT-contained serum can improve the PC12 survival rate and reduce the PC12 apoptosis induced by CORT. The mechanism of SZRT-contained serum on the calcium signal transduction path of PC12 cell apoptosis induced by CORT can be possibly attributed to the following aspects: Decreasing the expression of Caspase-3.
