Expression And Significance Of Survivin,Caspase-3 And NF-κBp65 In Colorectal Adenocarcinoma

Objectiv:To study the expression and significance of Survivin, Caspase-3and NF-kBp65 protein in tumorigenesis of colorectal adenocarcinoma.Methods:SABC immunohistochemical technique was used to examin the expression of Survivin,Caspase-3and NF-κBp65 at protein level in 45 cases of colorectal adenocarcinoma,40 cases of colorectal adenoma and 20 cases of normal colorectal mucosa.The level of the expression was analyzed combined with clinicopathological features of age and sex of the cases,the size and loci of the tumor,differentiation degree,histology type,lymphnode metastasis and Dukes-stages.Results:The positive rate of Survivin was 0%,35.0%and 64.4%in normal colorectal mucosare,colorectal adenoma and colorectal adenocarcinoma spectively, The expression of Survivin was increased in colorectal adenocarcinoma and has a significant difference with normal colorectal mucosare and colorectal adenoma (P＜0.05).The positive rate of Caspase-3 was95.0%,65.0%and 35.6%respectively in normal colorectal mucosa,colorectal adenoma and colorectal adenocarcinoma. The expression of Caspase-3 was decreased in colorectal adenocarcinoma and has a significant differences with normal colorectal mucosare and colorectal adenoma (P＜0.05).The positive rate of NF-κBp65was 5.0%,37.5%and66.7%respectively in normal colorectal mucosa,and colorectal adenoma colorectal adenocarcinoma. The expression of NF-κBp65 was increased in colorectal adenocarcinoma and has a significant difference with normal colorectal mucosare and colorectal adenoma (P＜0.05).The expression of Survivin and NF-κBp65 were higher in moderate and poor differentiation degree than those in high differentiation degree.The expression of Caspase-3 was lower in moderate and poor differentiation degree than those in high differentiation degree.The expression of Survivin was significant negative correlated to those of Caspase-3(P＜0.05).There were no significant differences in the sex of the cases,the size and loci of the tumor,lymph node metastasis and Dukes-stages of the expression of Survivin,NF-κBp65 and Caspase-3(P＞0.05).Conclusion:The expression of Survivin and NF-κBp65 were increased in colorectal adenocarcinoma.Survivin and NF-κBp65 may be closely correlated to the occurrence of colorectal adenocarcinoma.The Survivin and NF-κBp65 were increased belonged with the colorectal adenocarcinoma differentiation degree's depression.It presumed that the high expression of Survivin and NF-κBp65 may be the reflection of colorectal adenocarcinoma's malignant feature.The expression of Survivin in carcinoma was negative related to that of Caspase-3.It confered that survivin could prevented the apoptosis of tumor cell through restrained the activation of Caspase-3.There were no significant differences in the sex of the cases,the size and loci of the tumor,lymph node metastasis and Dukes-stages of the expression of Survivin,NF-κBp65 and Caspase-3.
