Objective : To study the distribution of p73 gene polymorphism in patients with colorectal tumor in Fujian Province, and explore the effect of p73 protein express on the development and progression in colorectal tumor.Method : All cases are from the patients who have received endoscopy and surgery in Fujian Provice Hospital.The samples from biopsy with endoscopy or surgery resection are confirmed colorectal adenocarcinoma or adenoma, the colorectal cancer is sporadic excluding Hereditary Non-Polyposis Colorectal Cancer(HNPCC). Recording patient's age, sex, susceptive factors to colorectal cancer in details. Blood sample acquitted from patients were divided into three groups, colorectal adenocarcinoma group (n= 60), adenoma group (n=60) and healthy volunteer group (n=60) respectively. Tissue sample acquitted from patients were divided into three groups, colorectal adenocarcinoma group (n=30), adenoma group (n=30) and the distant normal mucosa group (n=30) respectively. PCR--restriction fragment length polymorphism (RFLP) method was used to analyze p73 gene polymorphism. Immunohistochemistry staining method was used to show the expression status of p73 protein. Finally, the above results were tested by x2 and Fisher exact probabilities statistical analysis.Result:1.The frequencies of the p73 genotypes were 43.3% for wild-type (GC/ GC), 31.7% for heterozygotes (GC/AT) and 25% for variants (AT/AT) in colorectal adenocarcinoma patients,55.0 ,26.7and 18.3% in colorectal adenoma patients, 60.0,35.0 and 5.0% in normal control ,respectively. There were significant differences among colorectal adenocarcinoma, colorectal adenoma and normal control.(p<0.05) The polymorphism of p73 gene in relation to colorectal tumor. Apparently, the patients with colorectal tumor had a higher frequency of AT/AT ,comparing with that of the normal controls.The AT/AT homozygotes may have a greater risk of developing colorectal tumor.2.The GC/GC,GC/AT,AT/AT of p73gene polymorphism have no significant differences among patients who are different in age, sex, tumor location , tumor grading and Dukes'stage.(p>0.05)3.The expression of p73 protein.The case amount of p73 protein expression increase was 10 (43.3%) in colorectal adenocarcinoma tissues, 3 (10.0%) in colorectal adenoma tissues, 2 (6.7%) in normal tissues . The expression of p73 protein in colorectal adenocarcinoma tissues was significant higher than in colorectal adenoma tissues and in normal tissues .(p<0.05). The data show that the expression of p73 protein is in relation to development of colorectal adenocarcinoma cancer.4.There were no significant differences of p73 protein positive expression among patients who are different in age ,sex ,tumor location ,tumor grading and Dukes'stage(p>0.05).Conclusion:1. There were relationship between p73 gene polymorphism and colorectal tumor, the AT/AT homozygotes may have a greater risk of developing colorectal tumor in Fujian Province.2. Increase of p73 protein expression in relation to the development and progression of colorectal cancer.
