Objective:To observe the effects of all trans retinoic acid(ATRA) on the proliferation and adhesion of a human cervical cancer cell line:SiHa, and the effects of ATRA on the activity of MMP-2,MMP-9 enhanced by fibronectin(FN).Methods:Human cervical cancer cell line, SiHa,was treated by ATRA at different concentrations. Growth inhibition was measured by MTT assay, and cell adhesion assay was measured when SiHa cells treated by ATRA were incubated with FN or Mtrigel by MTT assay. Gelatin zymography was used to detect the actiity of MMP-2 and MMP-9 of SiHa cells incubated with FN at different concentrations and time point. The same way was used to detect the effects of ATRA at different concentrations on the activity of MMP-2 and MMP-9 enhanced by FN.Results:In this research we found that ATRA could inhibit the growth of SiHa at the concentration of 10-7mol/L,10-6mol/L,10-5mol/L, and the result showed that after 4 day of ATRA treatment, the growth of SiHa cells was inhibited obviously. The cells adhesion with FN was inhibited obviously when SiHa cells were treated by ATRA at the concentration of 8×10-6mol/L,10-5mol/L, and the cells adhesion with Mtrigel was inhibited obviously at 10-5mol/L ATRA. FN could induce the activity of MMP-2 and Pro-MMP-9 of SiHa cells with obvious activity effection when cells were treated with 5ug/ml FN(P<0.05). The activity enhanced little more when the concentration of FN raised(P>0.05). The activity weakened after 12h treatment. 10-7mol/L,5×10-6mol/L,10-6mol/L ATRA could obviously inhibit the activity of MMP-2 of SiHa cells enhanced by FN. and also can inhibit the activity of Pro-MMP-9 enhanced by FN.Conclusions:(1)ATRA can inhabit the growth of SiHa cells.(2)ATRA can inhibit the adhesion of SiHa cells.(3) FN can enhance the activity of MMP-2 and Pro-MMP-9 of SiHa cells.(4)ATRA can inhibit obviously the activity of MMP-2 of SiHa cells enhanced by FN, and can also inhibit the activity of Pro-MMP-9. |
