Study On The Active Fractions & Antitumor Activity Of Chinese Traditional Compound Medicine-WZDL

"Developing the effective fraction group of Chinese Traditional Compound Medicine(CTMC) is an available way to the modernization of Chinese herbal, we should carry on the serial research of Chinese herbal and CTMC at the basis of elucidating the curative action and mechanism." This is raised by relevant experts at "The Third Profound Scientific International Seminar of the Protection and utilization of Biodiversity". The effective fraction extracted from CTMC has been removed many components with adverse reaction, it's curative action is increased and adverse reaction decreased; chemical constituents are more clearly, can be easily formulate the quality standard and production.We studied the CTMC WZDL composed of five Chinese herbs, established the data bases of each herb (including resource, trait, property and flavor, meridian distribution, action and indication, chemical components and pharmacological activity in vitro and vivo). We extracted, isolated and purified four group of active fraction including lignanoids, saponins, tannins, polysaccharides, from Fructus Schisandrae Chinesis, Fructus Corni, Fructus Rosae Laevigate and Fructus Chebulae, Rhzoma Bletillae respectively.We compared several methods of extraction and isolation with the weight of dry extract, quantity of active consitituent, TLC identification, impurity test et al.. We found that the saponins could be isolated and purified more efficiently with macroporous adsorptive resins HPD-300, and subsequently got two active fractions(â…¢andâ…¤) with column chromatography from the original saponins which are eluted by 50 percent ethanol and 75 percent ethanol. We obtained the tannins(O) from the extraction of acetic ether; and two part ofpolysaccharides(â… andâ…¡) deposited with ethanol from the water-decocted liquid of the debris of four other herbs besides Rhzoma Bletillae; we also purified the lignanoids by soxhlet extraction with dissolvent of chloroform.We studied the anti-tumor and anti-adhesion activity of 0-â…¤active fraction and the compound on tumor cells in vitro. The results show thatâ…£andâ…¤have significant cytotoxicity and activity of anti-adhesion. The results of anti-turnor activity in vivo of compound WZDL show that it has inhibitory action on tumor cells, and the inhibition rate increased with dose.
