Alcoholic liver disease (ALD) is a public disease throughout the world with an ever-increasing number in the past decades. However, alcoholic minimal lesion and alcoholic fatty liver are completely cured by Abstinence and some treatments. It has been reported that phosphatidylcholine (PC) decreased when pathological changes occur in livers. So it may be a good way to cure liver disease to supply liver PC. In the study, we mainly focus on protective effect and mechanism of liver PC against alcoholic hepatic injury.(1)PC derived from swine liver was separated and purified by using Al2O3 column chromatography with 95% alcohol as eluent. The results show that liver PC was completely separately from PE by the elution with 95% alcohol, and its purity reached more than 90%. (2)Develop the model of alcoholic hepatocyte injury: hepatocytes are cultured with ethanol of different concentrations and time, the level of AST in medium increased while the survival rate gradually decreased, compared with those of the normal control group, with the consumption of ethanol increase. It shows that hepatocytes are damaged if the concentration of ethanol is higher than 100mmol/L and time is longer than 9 hours. (3)Study the effect of PC of different concentrations on survival rate and aminotransferase, and compare the effects of the soybean lipid, yelk lipid and polyene phosphatidylcholine on the MDA,GSH and TG of hepatocytes. It shows that 50nmol/ml PC can protect hepatocytes from injury. AST in medium,TG and MDA in hepatocytes increase but reduced GSH decreased. Our results suggest that liver PC play a role in the protective effects of hepatocyte against alcoholic injury. (4) Investigate the protective effects of liver PL and PC on ethanol-induced hepatic damage in mice. Compared with normal control group, the contents of the AST in liver injury group increased in serum, the contents of AST of low, middle and high dosage of PL groups decreased in serum, and the contents of GSH in high and middle dosage of PL group increased in liver tissue. Compared with liver injury group, the contents of TG is high dosage of PL group were decreased in serum. It is said that liver PL and PC have the protective effects on liver injury induced by ethanol in mice. All the results show that liver PC has the ability to protect from Alcoholic hepatic injury. It is related to anti-oxidative ability. PC influences the regeneration of hepatocytes and restrains hepatic injury.
