Relationship Between HPV16/18 Infection And Expression Of Ki-67, P16～(INK4a) And COX-2 In Invasive Squamous Cell Carcinoma Of The Cervix And Cervical In Traepithelial Neoplasia Of The Cervix Uteri

Objective: To study the expression and significance of HPV16/18, Ki-67, P16I^NK4a and Cox-2 proteins in invasive squamous cell carcinoma of the cervix(ISCC),cervical intraepithelial neoplasia(CIN). To investigate the correlation of HPV16/18 DNA and proteins referred to above. To analyse the significance of the development of ISCC so as to provide the valuable for the early clinical diagnosis and early therapy.Method: The sp immunohistochemical methods was performed to research the expression of Ki-67, P16^INK4a and Cox-2 proteins and in Situ. Hybridization was used to detect the expression of HPV16/18 DNA in 116 cases of cervical tissues.Results:(1) HPV16/18 in Situ Hybridization:The positive expression rate of HPV16/18 in ISCC, CIN and cervical condyloma acuminata was statistically significant compared to normal cervical tissue(P<0.05), and the positive rates from cervical condyloma acuminata, CIN to ISCC is upward trend.(2) The sp Immunohistochemistry results: ISCC, CIN compared to normal cervical tissue,the positive expression rate of Ki-67,P16^INK4a were significantly different(P<0.01).And compared with normal cervical, the positive rate of Ki-67, P16^INK4a from CINâ… , CINâ…¡-â…¢to ISCC is upward trend. In some normal cervical tissue and CINâ… , Ki-67, p16^INK4a have expression, but expression is weak and limited in quantity and expressed in the epithelial cells of the basal layer. The positive expression rate of Cox-2 protein in ISCC was significantly different(P<0. 01).(3) There were significant correlations between the expression of Ki-67 and HPV16/18; P16^INK4a and HPV(r=0.324, P<0.01; r=0.241, P<0.01). There was siginificant correlation between the expression of Cox-2 and HPV16/18(r=0.206, P<0.05),but is further explored to the specific mechnism.Conclusion: The expression of Ki-67, P16^INK4a in ISCC is siginificant correlated with HPV16/18 infection. They may be as valuable for the diagnosis of cervical cancer screening high-risk groups. Also suggested that cervical cancer high expression of Cox-2 and HPV16/18 have some relance.
