Objective: To observe the apoptosis in the rat Corpus Cavernous Penis after the bilateral cavernous nerve and/or dorsal nerve were transected.Method: 1.Animal model: Distribute the 78 SD rat randomly to 1,2,4,8,16,32d time point after the nerves were transected, each point is 13 ,there are group as dorsal nerve of penis,cavernous nerve and dorsal nerve and cavernous transection in per time point, each group is 3,and have3 for sham operation and 1 for normal control respectively. 2. Target detection: The qualition is detected by DNA Ladder, and the quantitation is detected by TUNEL. The sections of TUNEL are counterstained by hematoxylin, and the cell population of apoptotic nuclei and total nuclei are counted respectively (10 fields at x400 magnification per sample with 2 samples per group). 3. Location of apoptosis: The location of apoptosis is detected by dual stained with immunohistochemistry (with the antibody of actin-αand CD31 respectively) and TUNEL, the dual labeled cells are counted.Result: The variance of apoptosis ratio among different neruotomy group at per time point (except 32d time point) have significantly distinction (p<0.05) . The distinction of apoptosis ratio induced by cavernous nerve divided have statistical significanc compare with the dorsal never neruotomy,sham and control group respectively(p′<0.0046), the distinction between cavernous nerve neruotomy and cavernous nerve and dorsal nerve transaction combinately have no statistical dual stained (p′>0.0046);the have statistical significance of the apoptosis ratio between with the cavernous nerve and dorsal nerve transaction combinately group,the dorsal never neruotomy group,sham and control group respectively; The mean of dual stained cells is significantly different between in Smooth muscle and endotheliocyte (p<0.05).Conclusion: The injury of cavernous nerve can induce smooth muscle cells of corpus cavernous penis to apoptosis. The expression of apoptosis in corpus cavernous induced by dorsal nerve transected has no difference. Combination injury of cavernous nerve and dorsal nerve of penis can induce smooth muscle to apoptosis, and there no difference between with the only injury of cavernous nerve. Smooth muscle cells are the dominant cell population undergoing apoptosis.
