| ã€Purpose】To map the gene responsible for autosomal dominant high myopia ofchinese families by linkage analysisã€Methods】Four Chinese families(A,B,C,D) with high myopia were collected forthis study. A detailed clinical examination of the eye was performed for allpatients in all families. The genomic DNA of all members were extractedfrom peripheral blood leukocytes, according to the standard methods ofprotocol. D18s63 was selected from the MYP2 gene which harbor all knownlinked with high myopia. The marker were amplified by PCR. Fragmentswere separated by electrophoresis through 12% polyacrylamide gels.Haplotypes were constructed manually according to the pattern of the bandson the gels. Two-point LOD scores were calculated using the subroutineMlink of the Linkage package(version5.1).ã€Result】The maximum tow-point LOD scores was obtaint at D18s63, Zmax=1.36(θ=0.00) in family A, and other three families' tow-point LOD scoresare all bettwen-2 and +1.ã€Conclusion】The gene for autosomal dominant high myopia is linked D18s63 in family A, but we need more members to prove the linkage of D18s63 withthe gene for autosomal dominant high myopia in other three families。... |
