Background: Breast cancer is the most common malignancy in women. Several molecule markers are used in clinic which can help to make diagnosis and evaluate prognosis of breast cancer. However, the etiology of the majority of breast cancer cases is unknown. accordingly, it is important to study the molecular mechanisms of etiology of breast cancer and discover new gene markers. HOXD10 gene is one of the family members of homeobox genes. Some foreign scientists has found that HOXD10 gene is related to human breast cancer. They assume it is probably one of the inhibitors of breast cancer. In our country there are very few studies about the expression change in breast cancer. So in this study we have detected the expression of HOX10 gene in human breast cancer in China using Real-time RT-PCR method.Objective: To study the expression of HOXD10 gene in the primary human breast cancer and the correlationship with clinical pathologic factors.Methods: 1. 44 cases of primary breast cancer and 16 cases of benign breast lesion are reserved at -80℃. 2. Total RNA was extracted by trizol . The first-strand cDNA was acquired by reverse transcription . 3. The whole primer sequences were retrieved from gene bank of NCBI and the primers were designed by oligo 6.0 software. 4. HOXD10 and 18s rRNA genes segments which be used as standard substances of quantitation were amplificated by polymerase chain reaction. 5. Real-time reverse transcription polymerase chain reaction ( Real-time RT-PCR) was used to investigate HOXD10 mRNA expression on 44 cases of primary breast cancer and 16 cases of benign breast lesion. non-parametric test were applied to analysis the correlationship between HOXD10 expression and clinical pathologic factors.Results: 1. The slopes and correlation coefficients of the standard curves made by standard substances were fitted with criterion. 2. The expression range of HOXD10 mRNA of breast cancer is from 3.26 to 75 and the average value is 25.25. It is from 11.98 to 162.50 in benign breast lesion and the average value is 78.32. The expression of HOXD10 in breast cancer is decreased significantly compared with the expression in benign lesion. 3. A reduced mRNA expression of HOXD10 was examined in samples of histological grade III in breast cancer. 4. There were no significant correlations between expression of HOXD10 mRNA and clinical pathologic factors such as tumor size, lymphoid node matastasis and stage of tumor.Conclusion: 1. The expression of HOXD10 mRNA in breast cancer decreased significantly. And the expression of histological grade III in breast cancer was lower than grade I and grade II. It suggested that HOXD10 gene was probably one of the inhibitors of breast cancer and we should pay close attention to it. 2. In our research we did not find there were correlations between expression of HOXD10 mRNA and clinical pathologic factors. It maybe need verified again by another trials with more samples. 3. SYBR GREEN I real-time RT-PCR method which be used in our research have several benefits compared with the old one. It is not only a accurate method but also a simple way to detect the expression of genes.
