Objective:Aimed directly at the shortcoming in former work that rats'individual variation influencing experience results and semi-quantitation scoring method used to evaluate animal model less objectivity,adopted Open-field method to filt similar scoring rats entering next experience and evaluate the macroscopy appearance,providing objective and effective method for emotional disease animal model macroscopy evaluation;Aimed directly at the puzzle that macaques'social status distinguish method less fidelity and model evaluation scale checking index less satisfication,introduced the criterion to distin- guish social status in macaques and add the social subordination stress,improve fomer scale according to standard manufacture procedure;Investigated relationship of female experiment macaques'expression&behaviour and transmitter&hormone level in vivo by means of scale developed above offering experiment data and science explanation to dis- close modern mechanism of"Liver Losing Regulating".Methods:Adopted ethology and improved vaginal smear to determine rats oestrous cycle,apply open-field method to filt similar scoring rats entering experiment,adjust stimu- lus intensity and persistence time,introduce open-field method to estimate rats macroscopic appearance;Borrowed correlated reports abroad to standardize observation and discrimi- nation method of maca-ques,prepared Emotional Evaluation Scale about Female Experi- ment Macaque to estimate macroscopic appearance on macaques model.Results:In open-field experiment liver-qi invasion rats'scores were much higher than controlled while liver-qi depression rats'scores decreasing notably,the trend was retrieved after administration.About micro index detected on rats model liver-qi invasion rats'NE,DA,5-HT contents showed a increasing trend,while liver-qi depression rats DA content decreased and 5-HT content raised obviously in different encephalic region.Periphery monoamine neurotransmitter took on a opposite change compared with center.Liver-qi invasion rats E2,P contents showed a descending tendency as liver-qi depression rats had a similar change in serum in period of symptom presentation.It's just contradict in center. Scale after revising had a significant improvement,Test-Retest Reliability of the indigna- tion,depression,enjoyment and dread emotion were 0.90,0.92,0.95,0.92;Split-half Reliabi- lity were 0.96,0.79,0.87,0.97;Criterion-related validity coefficients were 0.93,0.74,0.61, 0.68.Norm of indignation, depression,enjoyment and dread factors were 21.35±5.13,17.96±4.88,12.68±2.26,21.25±4.47.It is found that liver-qi invasion macaques had a atypia negative correlation between 5-HT content and indignation emotion,while with the increas- ing of P content, the score of indignation factor descended rapidly in protophase and slow- ly in anaphase applying Scale method to investigate the relationship of expression&behav- iour and trans-mitter&hormone level in vivo on macaques model.Conclusion:Rats and macaques model analogued clinical manifestation and patholo- gical change satisfactorily.Modeling method was reliable and mature with a better credibi- lity and repeatability;Adopting open-field method could distinguish syndrome belongings of rats model clearly.Thus provided a direct evidence for differentiation of symptoms and signs on animal models. Scale had a satisfactory validity and reliability.It could disting- uish liver-qi invasion or depression macaques model explicitly.Based on above research work parts of the probable mechanism on"Liver Losing Regulating"were disclosed:Tant- rum appearance induced by Liver-qi regulating excite maybe related to NA,DA,5-HT level increasing in hypothalamus,limbic lobe;while depression symptom induced by Liver-qi regulating restrain maybe related to DA level descending but 5-HT level ascending in hypothalamus,limbic lobe.Supposed that"Liver Governing Regulating"may show its fun- ction through regulating these active substances and its possible function location is brain center,especially hypothalamus according to research above.
