Object: To investigate the different sensitivities of commonly used chemotherapeutic drugs on non-small-cell lung cancer(NSCLC) and gastric cancer by the methed of primary culture of NSCLC cells and gastric cancer cells,and to try to screen the best chemotherapeutic drugs or combined drugs.Methods: The drug sensitivity or resistance of 40 NSCLC samples to 9 commonly used chemotherapeutic drugs and combined chemotherapy were all detected by MTT colorimetric assay; The drug sensitivity or resistance of 34 gastric cancer samples to 7 commonly used chemotherapeutic drugs and combined chemotherapy were all detected by MTT colorimetric assay, and analyzed the results.Results: The individual chemosensitivity of NSCLC and gastric cancerwere comparatively different. About non-small-cell lung cancer(NSCLC): The average inhibition ratio of combined drugs was apparently superior to single drugs, P=0.038. The sequence for the average inhibition ratio of drugs was DDP+HCPT>DDP+VP 16>HCPT>DDP+NVB>DDP+ADM+IFO> ADM>CTX +ADM+VCR>DDP>NVB>VP16>CBP>IFO>VP16+CBP>VCR>CTX.The overall sensitivity of 9 chemotherapeutic drugs and 6 combined chemotherapy about NSCLC was different,P=0.000. There were significant difference in sensitivity among the single drug groups and among the combined drug groups, all P<0.01.About gastric cancer: The average inhibition ratio of combined drugs was apparently superior to single drugs, P<0.01. The sequence for the average inhibition ratio of drugs was 5-FU+MMC+ADM>5-FU+CF+VP16+DDP+ MMC>5-FU>5-FU+CF+VP 16>5-FU+CF+VP 16+DDP>5-FU+MMC+EPI>AD M>EPI>MMC>VP16>DDP>CF. The overall sensitivity of 7 chemotherapeutic drugs and 5 combined chemotherapy about gastric cancer was different,P=0.000. But there were no significant difference in sensitivity among the single drug groups and among the combined drug groups, all P>0.05.Conclusions: The MTT colorimetric assay was an exact, simple and rapid method for testing of chemosensitivity or resistance of anticancer drugs in vitro , and could give some guidances for clinic individual treatment and selecting reasonably chemotherapeutic scheme. The combined chemotherapy should be selected for NSCLC patients and gastric cancer patients.
