Object Probe the effects of immunologic tolerance induced by immature dendritic cells in bone marrow transplantation of H-2 haploidentical rat,researching new methods to prevention and cure immunological rejection in bone marrow transplantationMethods①adult male SD rats were made as donors,another adult hybridism female rats from filial generation of male SD rats and female Wistar rats were made as receptors,then building the bone marrow transplantation model of H-2 haploidentical rat.②peripheral blood mononuclear cells of SD rats were cultured with GM-CSF and IL-4 about 7 days,a great quantity of imDCs were harvested.③20 cases adult F1 rats were randomly divided into two groups as recipients:10 rats in experiment groups were enforced to BMT and imDC,10 rats in control group ware admined BMT without imDC infusion,and then did H-2 haploidentical BMT to observe the invasion status of acute graft versus host disease(aGVHD)Results①16 case survived in 40 days after BMT,the transplanted evidence(sex determining region of Y chrosmosome,SRY)was detected in 15 case.It showed we had builed BMT model successfully. During the derivation and cultivation of imDC from rat's PBMC, our everyday observations show:the morphous appearances of cultural cells were consistent with the characters of imDC.③the incidence rate of aGVHD in experiment group receptors was lower than that in control group receptors's(60%VS 100%).④the clinical aGVHD score of experiment group receptors was significantly lower than control group receptors's(2.7±1.0 VS 6.9±1.5).the Severity degree of Liver and Enteropathy pathology GVHD in experiment group receptors was notably lighter than control group receptors's.Conclusions①building in H-2 haploidentical rat bone marrow transplantation model and running pertinent researches was feasible by adult male SD rats and adult hybridism female rats from filial generation of male SD rats and female Wistar rats.②the imDC induced from PBMC in rat can be used by which studied on immunologic tolerance.③imDC can depress the incidence rate and the Severity degree of postgraft's aGVHD in in bone marrow transplantation of H-2 haploidentical rat.
