Objective: To examine the expression and correlation of Id-1 and VEGF in human colon cancer ,and study the roles of Id-1 in the progression and metastasis of human colon cancer. To transfect the positive and negative eukaryotic expression vector of Id1 to the SW480 cell lines of colon cancer ,and investigate the relationship between Id1 ,VEGF expression and cell proliferation , apoptosis in colon cancer cell lines,and provide experimental evidence to the targeted therapy of colon cancer.Method:1. Expression of Id1 and VEGF proteins was detected by S-P immunohistochemical technique in colon carcinoma and the adjacent normal colon tissues ;2. To transfect the positive and negative eukaryotic expression vector of Id1 to the SW480 cell lines of colon cancer , and successfully select the stabilized expressed cells lines. to detect the expressions of Id1 and VEGF proteins and mRNAs by semi-quantitative RT-PCR and S-P immunocytochemistry technique; Cell growth curve was obtained by MTT assay to emanine the cell apoptosis and proliferation .Results:1.The postive rate of Id1 and VEGF protein was significantly higher in colon cancer than in normal tissues (P < 0.05 ), and its expression have the positive correlation.2. Successfully transfect three types of vector to the SW480 cell lines of colon cancer and finally select the stabilized expressed cells lines. The result display of Id1and VEGF expression by semi-quantitative RT-PCR and S-P immunocytochemistry technique is that , the groop of transfecting the positive eukaryotic expression vector of Id1 (Abbr. PId1) has the higher expression compare to the control group on the lever of proteins and mRNAs, and the groop of transfecting the negative eukaryotic expression vector (Abbr. NId1) of Id1 lower expression significantly than the control group .3. the Cell growth curve by cell counter and MTT assay show that the pace of proliferation of NId1groop is lower than the control group(P < 0.05),and the PId1 groop is near to the control group(P>0.05).Conclusion: 1. The Id1 protein expression was higher significantly in colon cancer than in normal tissues, means that its expression has an important relation with the progression of human colon cancer; and the expression level of Id1 was related to extent of infiltrating,that histological differentiation and Dukes stage( P < 0.005),and was not related to lymphnode metastasis,sexual differenation and age( P > 0.05).2. After transfecting the negative eukaryotic expression vector of Id1 to the SW480 cell lines of colon cancer , the Id1 protein and the VEGF protein expression reduces, cell multiplication ability drops, but if transfers the positive eukaryotic expression vector of Id1 to the SW480 cell lines of colon cancer ,the function is opposite, prompts may Id-1 take a treatment target spot to treat the colon cancer, and may through reduce the expression of VEGF which is one method of controlling tumor metastasis. |
