Objection: glutathione(GSH) is a tripeptide(L-γ-glutamyl-L- cysteinyl-glycine) that serves several essential functions within the cell, It is the most abundant nonprotein thiol in almost all aerobic species, occurring at intracellular concentrations of 0.5 to 10 mM.. GSH is low-molecular-weight, water-soluble and can easily traverse between membrane,capillary wall,all kinds of biomembrane and blood-cerebral barrier, it is a main determinate facter in cellular redox state, a intemal radical scavenger. By comparing of serum oxyradical and antioxygen level, nitrogen monoxidum activity, clinical symptom, electrocardiogram appearance, contraction and diastolic function between convetional therapy and glutathione treatment in patients with coronary heart disease, investigate correlation ship between oxyradical level, nitrogen monoxidum activity and clinical symptom, cardiac function, to research the therapeutical effect of glutathione on coronary heart disease and provide its mechanisms, evaluated its relative therapeutic values.Method: eighty-five subjects with diagnosis of coronary heart disease were selected in hospital patients, 45 male and 40 female. All these diseases should be excepted: uncontrolled hypertension, periphery vascular disease, chronic cardiac failure, thyroid disease, diabetes, hepatic and renal disease, inflammation, tumor and other serious disease, and have not received any antioxidants or statins for two weeks. the patients were divided into two groups: 1)therapeutic group: convetional treatment and with glutathione 240mg intravenous drip, once a day, for 14 days. 2)contrast group: conventional treatement, the MDA, SOD, NO levels in serum were measured, electrocardiography ( ST stage, T wave) and echocardiography(CO, EF,FS,IVRT,Ei,Ai) was examined pretreatment and posttreatment each groops. Record the data mentioned above and manage statistic analysis with SPSS 12.0.Result:1. the levels of MDA, SOD, NO in serum changed in each groops posttreatment (P<0.01), and the TG groop has differed significantly from CG (P<0.05, P<0.01)2. there are significantly correlation ship between the improvement of redox state and improvement of symptom and ECG.3. glutathione can improve ventricular diastolic function IVRT, Ei (P<0.05), has no effect on contraction function.4. there is significantly correlation ship between the improvement of ventricular diastolic function and improvement of symptom(P<0.05,, P<0.01).Conculsion: The results indicate that glutathione can restore redox state, improve the function of endothelial cell, control symptom and improve ventricular diastolic function in patients with coronary heart disease, the mechanism is related to its antioxidation function.
