Objective: To establish the rats model to study nephrorrhagia with gross hematuria.Methods: SD rats weighing 350~400g were randomly divided into 2 groups (6 rats in each group): puncture group and normal control group(sham puncture group).All rats were treated by a serie of operation :intra-ureteral cannula+whole body heparinization+diureses+fluid replacement.The rats in puncture group were punctured by renal biopsy needle on the same side kidney to establish the models of nephrorrhagia with gross hematuria,while the rats in sham puncture group had not been punctured.Systolic blood pressure(SBP)and 2-hour mortality were inspected and noted ,meanwhile such lab index: urinary red cell, blood pigment and serum creatinine were detected in all rats before, 1 hour and 2 hour after renal puncture. Finally,deal with the data statistically.Results: After punctured by biopsy needle on the kidney, there were clear gross hematuria flowing out from the ureteral catheter. The incidence of gross hematuria in puncture group was 100% and 2-hour mortality was 33.33%,while no rat in sham puncture group had gross hematuria or died (P <0.05).SBP and the level of blood pigment were rapidly decreased significantly than the sham puncture group(P <0.05).Conclusion: We succeed in setting up the rat models of nephrorrhagia with gross hematuria by kidney puncture+whole body heparinization. It was an effective and reliable animal model with good repetitiveness,which can collect the lab specimen directly to assay some index reflecting renal hemorrhage. Objective: To investigate haemostasis effect of retroperfusion thrombase through ureteral catheter on nephrorrhagia rats.Methods: After the rat models of nephrorrhagia with gross hematuria had been established by kidney puncture+whole body heparinization,32 SD rats weighing 350~400g were randomly divided into 4 groups (8 rats in each group): low-dose thrombase group(125 U/ml), middling-dose thrombase group(250 U/ml) , high-dose thrombase group(500u/ml)and normal control group(equal capacity NS).All rats were retroperfused by three different doses of thrombase or NS through ureteral catheter respectively.Systolic blood pressure(SBP) and 2-hour mortality were inspected and noted ,meanwhile such lab index: Urinary red cell, blood pigment, serum creatinine,bleeding time(BT) and clotting time(CT) were detected in All rats before, 40min,2hour after retroperfusion. Finally,deal with the data statistically.Results:①Among 4 groups,the difference of level of Urinary red cell , blood pigment and serum creatinine was not significant(P >0.05) after nephrorrhagia model was established(before retroperfusion).②Compared with normal control group,the level of blood pigment and SBP of the other 3 groups was higher respectively(P >0.05),while the level of red blood cell count in urine was lower(P >0.05).The level of above-mentioned index among high-dose group ( 500U/ml ) and middling-dose group(250 U/ml)versus low-dose group was significantly different(P <0.05),while the difference between high-dose group(500 U/ml)and middling-dose group(250 U/ml)was not significant 5min and 40min after retroperfusion respectively(P >0.05).③Among 4 groups ,the difference of bleeding time(BT),clotting time(CT) and the level of serum creatinine before versus 40 Min after retroperfusion was not significant (P >0.O5)。Conclusion: All three different doses of thrombase had haemostasis effect on renal hemorrhage rats by retroperfusion through ureteral catheter,and the effects were related to the medicament and the dosage;while retroperfusion thrombase did not effect BT,CT and the level of serum creatinine of rats.
