Objective:To observe the expression and significance of Keratinocyte Growth Factor(KGF) and its receptor(KGFR) in cholesteatoma tissues. To investigate the role and significance of KGF,KGFR in the epithelial hyperproliferation and development of cholesteatoma. The labeling index for Ki-67 was used to assess the proliferative activity and to examine the hyperproliferate state of keratinocyte in cholesteatoma tissues.Method:We examined the expression of KGF,KGFR and Ki-67 by immunohistochemical staining (SP) in 45 samples from middle ear cholesteatoma and 15 epithelial cells samples from the external auditory meatus.Result:1. The results showed that KGF in cholesteatoma tissues (91.11%) was higher than that in normal skin epithelial tissues (46.67%), the expression level of KGF in cholesteatoma tissues was significantly increased (P<0.01).2. The results showed that KGFR in cholesteatoma epithelial tissues (88.89%) was higher than that in normal skin epithelial tissues (40.00%), the expression level of KGFR in cholesteatoma epithelial tissues was significantly increased (P<0.01).3. The results showed that Ki-67 in cholesteatoma tissues (64.44%) was higher than that in normal skin epithelial tissues (20.00%), the expression level of Ki-67 in cholesteatoma tissues was significantly increased (P<0.01).4. The results showed Ki-67 was significantly higher in KGF+/KGFR+ cholesteatoma tissues than other cases. The expression of KGF and KGFR, KGF+/KGFR+ and Ki-67 was present positive correlation in cholesteatoma tissues (P<0.05). Conclusion:The expression of KGF,KGFR and Ki-67 in cholesteatoma tissues was significantly higher than that in normal skin epithelial tissues. The higher expression of KGF in cholesteatoma tissues explained epithelial cell possessing strongly reproductive activety. Because epithelial cell possessed strongly reproductive activety, precipitating epithelial hyperproliferation and also verificating it was involved in signal transmission between mesenchymal cells and epithelial cells. There was present positive correlation between KGF and KGFR in cholesteatoma tissues which verificated the paracrine action between KGF and KGFR. KGF exerted its biological function in the epithelial cells of cholesteatoma tissues by specific binding KGFR. Then enhanced the epithelial hyperproliferation of cholesteatoma tissues. The higher expression of Ki-67 indicated that many epithelial cells were occupied in cell generation cycle in cholesteatoma tissues. And it reflected the proliferation status of cholesteatoma epithelium. In statistics there was significantly higher proliferative activity in cholesteatoma tissues than normal skin specimens. We found that cholesteatoma expressing KGF, KGFR and Ki-67 highly, which proliferative activity was also higher. These results strongly indicated that KGF and KGFR were closely correlation with the epithelial hyperproliferation and development of cholesteatoma tissues.
