Objectives 1.To detect Apolipoprotein A5 (ApoA5) polymorphism by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP). 2. To study the APoA5 genotypes and its alleles frequencies distribution of normal Chinese of Han people from Anhui province. 3. To investigate the associativity between APoA5 polymorphism and the serum levels of the lipid and lipoproteins.Methods 1. To collect 156 normal Chinese without blood relation of Han people from Anhui province. To design the enzyme digestion site for restriction endonuclease MseI around -1131 according to the promoter region of ApoA5 gene. To detect the genetype as well as the serum levels of total cholesterol(TC), triglyceride(TG), low density lipoprotein(LDL), high density lipoprotein cholesterol(HDL), and very low density lipoprotein(VLDL) with the PCR-RFLP. 2. To calculate the ApoA5-1131T>C genotypes and its alleles frequencies distribution. 3. To analyze associativity between APoA5-1131T> C polymorphism and the serum levels of the lipid and lipoproteins from Anhui province.Results: The frequency distribution of APoA5 genotypes of the normal Chinese of Han people from Anhui province was: TT:53.21%,TC=30.12%,CC=16.67% respectively, and the frequency distribution of the alleles was T=0.68,C=0.32. The frequencies distribution of APoA5 genotypes and their alleles from Anhui province were similar to who were from the other area of China and Japan, but there was a significant difference between them and the people who were from India and England(P<0.05, P<0.01). The blood levels of triglyceride(TG)in TC and CC genotypes were significantly higher than those in TT type(P<0.05),especially the serum level of TG which in CC type was the highest, but there was no significant statistical difference between the TG in CC type and which in TC type(P>0.05).The CC type carriers (TC+CC)had a significant increase in TG levels compared with non-c carriers (TT) (P<0.05), and the blood levels of TC, LDL,VLDL, HDL-C had no significant difference between the two groups. The female C carries had a significant statistical difference in TG levels compared with non-C carries(P<0.01),meanwhile the serum level of TG in male C carries was higher than those in non-C carries, but no significant statistical difference( P>0.05)。Conclusion: The frequency distribution of APoA5-1131T> C alleles of the normal Chinese of Han people from Anhui province is similar to those from the other area of China and Japan, but there is significant difference between them and the people who were from India and England. 2. There is an associativity between APoA5-1131T> C polymorphism and the serum level of TG, but no associativity between APoA5 polymorphism and the serum levels of TC, LDL, VLDL, HDL-C. 3. The normal Chinese of Han people in Anhui province who carries ApoA5-1131 C alleles are higher than the others, and it is associated with the increased serum level of TG, especially in females.
