primary part The Investigation of Serum 8-Isoprostane, GSH-Px inPatients with Obstructive Sleep Apnea/hypopnea SyndromeObjective To investigate the expression of Serum 8-isoprostane, glutathioneperoxidase(GSH-Px) in patients with obstructive sleep apnea/hypopnea syndrome(OSAHS), and explore changes of oxidative stress in patients of OSAHS in vivo.Methods 15 patients with simple snoring and 65 patients with OSAHS wereinternalized in the study. After being collected common clinical data, all patientsunderwent a single:night polysomnogram to get polysomnogram variables. Serum8-isoprostane was measured by enzyme-linked immunosorbent assay (ELISA), and serumglutathione peroxidase(GSH-Px) was measured by spectrophotometer.Result 1. The levels of 8-isoprostane increase gradually with the increase of apneahypopnea index(AHI), while the levels of GSH-Px decrease gradually with the increase ofAHI. 2. 8-Isoprostane correlates with AHI, oxygen desaturation index(ODI), cumulativepercentage of sleep time with SaO2 below 90% (CT90%), average SaO2, minimum SaO2,Epworth sleepiness scale(ESS), body mass index(BMI) respectively, meanwhile,correlation coefficient(r) decreased gradually from AHI to BMI. 3. GSH-Px also correlateswith ODI, AHI, average SaO2, CT90%, ESS, minimum SaO2, BMI respectively, and rdecreased gradually from ODIto BMI.Conclusion 1. The severity of OSAHS is independently associated with oxidativestress. 2. Serum8-isoprostane, fasting insulin(FINS) were measured by enzyme-linked immunosorbentassay(ELISA), and serum glutathione peroxidase(GSH-Px) was measured byspectrophotometer. HOMA-IR, FINS were used to estimate insulin resistance(IR),HOMA-βwas used to estimate islet beta Cell function.Result 1. With the increase of apnea hypopnea index(AHI), serum fasting bloodglocuse(FBG), FINS, 8-isoprostane, GSH-Px are significantly different among the snoringgroup, mild-moderate OSAHS group and severe group(P=0.0034, P<0.0001), butHOMA-βdoes not differ significantly among the three groups(P>O.05). 2. Serum FINSand HOMA-IR correlate with AHI, 8-isoprostane, GSH-Px, BMI respectively(P<0.05).When adjusted for age and BMI with multiple linear regression analysis, FINS andHOMA-IR correlate with AHI, 8-isoprostane respectively as before. There are nocorrelation between HOMA-βand AHI, 8-isoprostane, GSH-Px, BMI(P>0.05). 3. SerumFINS, HOMA-IR correlate with ODI, CT90%, minimum O2, average SaO2 respectively,with correlation coefficient(r) decreased gradually from ODI to average SaO2 FINS andHOMA-IR correlate with ODI, CT90%, minimum SaO2 respectively. When adjusted for age and BMI with multiple linear regression analysis, multiple correlation coefficient(R)decreased gradually from ODI, CT90% to minimum SaO2.Conclusion 1. Augmented oxidative stress and distinctive intermittent hypoxia ofOSAHS correlate with IR independently. 2. IR correlates with frequency of desaturationmore strongly than with severity of desaturation. |
