Objective: On the basis of subacute decrepitmodel established. two kinds of mulriplc model through injecting Aβ(25)-(35) to brain and irrigating aluminium to stomach for rats.And at the aspect of respetive behavior,biochemical check-up and histology progress contrast and reseach.Methods: Adopting injected D-Galactose peritoneally for six weeks establish subacute model of decrepitude of wistar rats.on the base of it .establish the old dementia through injecting Aβ(25)-(35) to hippocampus andirrigating the aluminium to stomach.Behavior test ability of study ang memory in rats.through the water maze experiment and jumping stand experiment.Biochemistry test the vigor of cholinesterase and hippocampus tissue .Histology progress HE dyeing and argentums dyeing which utilize photic microscope.and observe pathological variety of hippocampus tissue of rats which utilize transmission microsope.Results: (1)Study of behavior experiment show that compared with the D-galactorse group.the ability of memory in Aβ(25)-(35) model group and aluminium toxicosis group descended obviously. (2)Biochemistry experiment show that compared with the D-galactorse group,the vigor of cholinesterase and content of acetylcholine in brain cortex and hippocampus Aβ(25)-(35) model group and aluiminium to xicosis group descended obviously but for the vigor of cholinesterase.there is not different evidrently number be between the two model group.(3) Histology groory HE dyeing show that the nervecells in brain cortex descended for the two kinds of mulriple model group.and the necrotic nervecells concentrated.Especially in the Aβ(25)-(35) model group.it can be observed that the nervecells of syknosis,the brain cortex which dyed deeply, and it is pyknosis.Argentum clyding show, there have senile plaques (sp)in brain cortex for Aβ(25)-(35) model group. Transmission show nerve cells in the hippocampus.in some extent.appear to recessive variety.nucleus concentrated, lysosome and lipofuscin increased.mitochondrion tumefy.crest parted and caritated Synapse's number in the nerval felt descended,a part of axoplasm in the axone cavitated.mitochondrion cavitated.above phenolmenon. Aβ(25)-(35)model group was best evidence.Conleusion: on the basis of D-galactose worlced subacute decrepit model.The two levels of mulriple moldel which Aβ(25)-(35) model and aluminium toxicosis model can be established.At the aspect of AD pashtolgy histological variety.Aβ(25)-(35) modle group excelled aluminium.toxicosis group.through statisxics's analysis for the content of acety lcholine,aluminium to xicosis group excelled Aβ(25)-(35)model group.There for, The two kinds of AD multiple model nosonly can be used to basel research of the old dementia,but also can be used to investigate the prevention and cure of the old dementia. |
