| adhesion molecules are kinds of glucoproteins residing in cell surfhce, mediating adherent and contiguity effect among cell to cell and cell to extracellular matrix. They are various and widespread distributed. Inter cellular adhesion molecule-1(ICAM-1) is generated in endotheliocyte,and contacts with natural ligands(two members of leukocyte integrin CD18 family), which are lymphocyte function associated antigen-1(LFA-1) and macrophage differentiation antigen-1(Mac-1), resulting in blood leukocyte immigrating blood vessel endothelium and reinforcing adhesion between monocaryon and endothelial cell, encouraging releasing of interleukin-1(IL-1), tumor necrosis factor -α(TNF-α) interferon-r(IFN-r), priming artherosclerosis and making advancement, so it's expression level and changing activity take significant rolein vascular lesions. In recent studies, ICAM-1 the 469-th amino acids in the sixth exron heterogenesis(A G), which polymorphism has connection with the change of the molecular activity and the generation of vascular lesions.ObjectivesThis study analyzes the sixth exon K469E polymorphism in ICAM-1 gene through detecting this polymorphism distribution frequency in 374 han race patients with Type 2 diabetes in Tianjin area and 139 normal persons, and analyzes the dependablity with nosogenesis of diabetes and the micrangium complications.MethodsFasting blood sample of 374 T2DM patients and 139 health controls who were unrelative were collected randomly, the genome DNA was obtained from the leukocytes, and then PCR-RLFP method was performed to investigate the genotype of ICAM-1 gene the sixth exon the 469-th amino acid cite.Results1 A, G alleles and AA, AG, GG genotypes were found in ICAM-1K469E gene cite in T2DM subjects, From the statistic data, we found that the frequency of AG GG genotypes and G allele are significantly increased in T2DM than in normal control group.2 In the analysis of K469E polymorphism in different phases od DN, There is a trend that the more serious the DN(DN-0, DN-1, DN-2) is, the more frequency of G allele and AG+GG genotypes rise。and both have statisticant difference.3 A decreased frequency of G alleles and AG+GG genotypes inT2DM with DR(diabetes retinopathy), but there is no significant difference.4 General analysis of T2DM with nephropathy and retinopathy shows that the lowest frequency of G alleles and AG+GG genotypes in the group of T2DM samole with retinopathy, the highest frequency in the group of T2DM with nephropathy and the frequency of those in the group of T2DM with both complications was similar with that of without the two complications. And there are no significant difference.Conelusions1 The frequency of G alleles and AG+GG genotypes were found significantly increased in ICAM-1K469E gene cite in T2DM subjects, maybe they are the predisposing genes of T2DM.2 The frequency of G alleles and AG+GG genotypes were found significantly increased in T2DM with nephropathy subjects, and G allele maybe take part in the cause and severity of DN.3 The frequency of G alleles and AG+GG genotypes were found relatively decreased in DR than without complications, which need the further study of a great quantity of samples. |
