Da-Xuan-Ji-Wei is the whole herb of Onychium japonicum (Thunb.) Kze ofOnychium genus, which is distributed mainly in Japan, Korea and other Asiaticcountries. In China it is distributed mainly in the south of Qinling Mountains, thenorth of Guangdong, Shanxi and Taiwan province. Onychium japonicum has theeffects of clearing heat, removing damplaw, arresting bleeding, hemostasis, and isused for treatment of wind-heat type common cold, acute gastroenteritis, dysentery,jaundice, hemoptysis, hematemesis, hemafecia, hematuria, furunculosis, injury andpain in traditional Chinese medicine.The material (Onychium japonicum) in this study is the folk medicine of TujiaNationality in Western Hubei, which has common use and remarkable effects intraditional Tujia Nationality medicine, on the basis of the investigation about the folkmedical plants in Tujia Nationality of Hubei. We began to study on the bioactiveconstituents from O. japonicum, in order to develop modem drug from this traditionalChinese medicine.By using repeated column chromatography, including Silica gel, Sephadex LH-20,Toyopearl HW-40 and preparative HPLC, 31 compounds were isolated from the P.E.and EtoAc extracts of the Onychium japonicum. By the means of IR, UV, MS,1H-NMR, 13C-NMR, 1H-1HCOSY, 13C-1HCOSY, HMBC, NOESY spectra, opticalrotation and chemical reaction. The structures of eight new compounds, one newnatural product and ten known compounds were identified. Eight new compounds,conclude flavonoid, chalcone and coumarin types of structure, named Japonicone A-F,Japonilactone A and B. The known compounds are eriodictyol, luteoloside, butin,protocatechuic acid, dihydroxy-acetophenone, caffeic acid, vanillic acid, p-hydroxysalicylal, 4, 6-dimethylgallate andβ-sitosterol.The in vitro antitumor activity of isolated components from the P.E., EtoAc and BuOH extracts was tested. Compound 1 exhibites the positive cytotoxic activityagainst the human carcinoma cell lines Hela and BEL-7402 in vitro. The inhibitionrate of Hela cell is 41.49% (30μg/ml), and Bel-7402 cell is 45.87% (30μg/ml). |
