Objective: The purpose of this study was to investigate the the morphological changes and expression of NGF Receptors: p75 and trka in lumbar dorsal root ganglia(DRG) and bladder of streptozocin-induced diabetic rats. At the same time , the present study was aimed at clarifying these factor's roles in diabetic cystopathy(DC), and supply experimented data for the clinical and fundamental study on DC.Methods: Experimental animal was Sprague-Dawley(SD) female rats, provided by Hebei Medical College Experimental Animal Center.35 Female SD rats was divided into 2 groups stochastically, control group 15, diabetes model group 20,and the body weight was from 200 to 250g. diabetic rats model was indused by using the STZ single abdominal cavity injection and the dosage was 60mg/ kg, isochoric citric acid buffer solutions for control group .Two groups were given the ordinary animal feeds that were bought from Hebei Medical College Experimental Animal Center. Blood glucose (blood glucose,BG) was measured after injecting the STZ for 72 hours . Diabetic model was successfully built when BG>16.7mmo1/L. Two group of animal tail venous blood glucose were measured when 8 weeks after diabetic model is successfully built, those rats whose blood glucose was lower than 16.7mmol/L was removed.Experiment was carried out when 8 weeks after diabetic model was built and before the experiment metabolism cage 24 hours amounts of urine was measured . Under 5% chloral hydrate anaesthesia, after whole body irrigation using physiological saline and 4% paraformaldehyde phosphoric acid buffer solution (pH7.4) that containing 0.1%DEPC respectively, whole urinary bladder and both sides of L6S1 DRG were cut off . Urinary bladder wet weight was measured and all specimens were fixed immediately. Using Haematoxylin - Iraq red (HE) dyes, morphological change of diabetes rats urinary bladder detrusor was observed under microscope.Experimental DC model was confirmed based on two group of rat's blood glucose, body weight, urinary bladder wet weight, 24 hour amount of urine change and the morphological change. Trka and p75 expression in bladder and DRG were measured by immunohistochemical staining .Under microscope the results were observed, it was positive result in the urinary bladder detrusor cell or the DRG neuron when yellowish brown pellet emerges .From each slice 8-10 fields of vision were chosen and 100 cells were registered to abserve morphological change under the high objective lens.
