Objective:The purpose of this study is to establish the expression of NF-κB,caspase-3 and the apoptosis states in middle ear cholesteatoma,discuss the relationship of NF-κB,caspase-3 and apoptosis states in middle ear cholesteatoma,explore the possible role of NF-κB,caspase-3 in middle ear cholesteatoma.Let us know pathogenesis ofmiddle ear cholesteatoma deeply.Methods:25 cases of cholesteatoma and 10 cases of normal external meatal skin were obtained from middle ear surgery.1.The expression of caspase-3 and NF-κB in middle ear cholesteatoma were investigated by immunohistochemitry methods.2.The apoptosis states of the 2 tissues were determined with TUNEL methods To discuss the relationship of caspase-3,NF-κB and the apoptosis states in middle ear cholesteatoma.Results:1 Comparing with normal meatal skin,the expression of NF-κB protein significant increased in cholesteatoma.The positive unit(PU)is 27.90±9.13,13.23±9.05(t=4.307,P<0.05) respectively.The inflammatory cell and vascular endothelial cells in the paramatrix also showed positive expression.The PU of caspase-3 protein expression in the epithelium of middle ear cholesteatoma tissue and normal skin were 17.29±6.21,8.31±3.44(t=4.287,P<0.05)respectivety. 2.The apoptosis of cholesteatoma tissue were higher than that in normal meatat skin.The PU is 15.51±6.73,7.79±3.76(t=3.398,P<0.05)respectively.3.Statistical correlations were established between apopotosis of the cholesteatoma and the caspase-3 protein.Conclusions:Comparing with normal meatal skin,the expression of NF-κB protein significant increased in cholesteatoma.It show us that NF-κB play a key role in the occurrence and development of cholesteatoma.The apoptosis of cholesteatoma tissue were higher than that in normal meatal skin,and caspase-3 protein is higher expression in cholesteatoma than normal meatal skin.It suggest apoptosis play an important role in the pathogenesis of cholesteatoma.
