Objective:To study the effect of aspirin preconditioning to the content of IL-6 and TNF-αin brain tissue after focal cerebral ischemia reperfusion in rats.To study the neuroprotective mechanism of aspirin preconditioning to the inflammation in focal cerebral ischemia reperfusion and its optimal dose.Methods:Sixty healthy male SD rats were randomly divided into five groups:sham-operation group,ischemia control group,aspirin preconditioning 10mg/kg group,aspirin preconditioning 50mg/kg group and aspirin preconditioning 150mg/kg group,12 rats in each group.The aspirin preconditioning groups were taken suspl of aspirin tablet by intragastric administration five days before operation,one time per day.The other two groups were taken normal saline by intragastric administration five days before operation,one time per day.The middle cerebral artery occlusion(MCAO)model was made by the suture method.After middle cerebral artery occlusion for 2 hours and reperfusion 24 hours,the content of TNF-αand IL-6 of serum and the brain homogenate by RIA method,the infarct volume and the neurologic deficit were tested.Results:1.In the low dose and middle dose of ASA preconditioning groups,scores of neurological deficit and infarct volume were low compared with the model group(P<0.01).2 In the low dose and middle dose of ASA preconditioning group,levels of the brain homogenate TNF-αshow significant differences compared with the model group(P<0.01).3 In the low dose of ASA preconditioning group,levels of the brain homogenate IL-6 shows significant differences compared with the model group(P<0.01),levels of the brain homogenate IL-6 shows statistical differences compared with the model group in the middle doses of ASA preconditioning group(P<0.05).Conclusion:1ASA preconditioning can lessen the neurologic deficit after the focal cerebral ischemia reperfusion.2 ASA preconditioning can minify the infarct volume after the focal cerebral ischemia reperfusion.3 ASA preconditioning can inhibit the inflammation in the process of the focal cerebral ischemia reperfusion,and its mechanism is related to the inhibition of TNF-αand the heightening of IL-6.4 The low and middle doses of ASA preconditioning works well in lessening the inflammation in the focal ischemia reperfusion. |
