ObjectiveTo establish rabbit high cholesterol atherosclerosis model and observe the expression of angiotensinⅡ(AngⅡ)and chymase in myocardium,investigate the influence of Perindopril Valsartan and Atorvastatin to the expression of angiotensinⅡ(AngⅡ)in myocardium.MethodsThirty-eight healthy male New-Zealand albino rabbit were divided into five groups randomly:(Group A)Normal control(n=6);(Group B)high cholesterol diet(n=8);(Group C) high cholesterol diet and Perindopril(0.5mg/kg/day)(n=8);(Group D)high cholesterol diet and Valsartan(10mg/kg/day)(n=8);(Group E)high cholesterol diet and Atorvastatin(2.5 mg/kg/day)(n=8).After feeding for eight weeks,Collecting the empty stomach blood preparation through central artery at awake state at the end of eight weeks(fasting diet twelve hours),centrifugalization(3000rpm,10min),blood serum was stored at -20℃and evaluate the density of the total cholesterol(TC)and low density lipoprotein(LDL)by BackmaanLX20 automatic biochemistry analyzer.To observe Arteriae aorta by morphology observation and HE dyeing.To determine the expression of AngⅡby immunohistochemistry and the results were demonstrated by Positive Unit(PU).To determine the expression of chymase by Western blot and the results were demonstrated by gray scale.Result1)The AngⅡexpression of high cholesterol diet Group higher than Normal control Group (21.65±4.20 vs 4.58±1.20,P<0.05);The AngⅡexpression of high cholesterol diet and Perindopril Group lower than high cholesterol diet Group(13.00±1.77 vs 21.65±4.20,P<0.05), but higher than Normal control Group(13.00±1.77vs4.58±1.20,P<0.05);The AngⅡexpression of high cholesterol diet and Valsartan Group have no difference with high cholesterol diet Group (22.30±1.58 vs 21.65±4.20,P>0.05),but higher than Normal control Group(22.30±1.58 vs 4.58±1.20,P<0.05);The AngⅡexpression of high cholesterol diet and Atorvastatin Group lower than high cholesterol diet Group(16.50±1.53 vs 21.65±4.20,P<0.05),but higher than Normal control Group(16.50±1.53 vs 4.58±1.20,P<0.05)。2)The chymase expression of high cholesterol diet Group higher than Normal control Group(30.88±4.05 vs 177.17±6.18,P<0.05); Conclution1)The expression of AngⅡand chymase are higher in cholesterol diet Group than in Normal control Group;That explain myocardium RAAS is at the state of activation when AS taking place.2)Perindopril and Atorvastatin can cut down the myocardium AngⅡexpression of rabbit atherosclerosis model,but Valsartan can't.
