Objective:study on the method of Human mammary cancers primary culture in vitor,using FCM and MTT assay we detected the individual sensitivity of primary culture cell of mammary cancer to ADM,MTX,TAX,5-FU,MMC and VCR finding a way to prediction chemosensitivities.Methods:(1)primary Human mammary cancer were obtained from human body in operating room and cultured in vitro.(2)The cultured mammary cancers cell were divided randomly into 7 groups:the normal control,ADM,MTX,TAX,5-FU,MMC and VCR treatment 1,2,3,4,5,6 group. 1,2,3,4,5,6 group were stimulated by ADM,MTX,TAX,5-FU,MMC and VCR in 5PPC for 48 hours,the normal control group were incubated with equal conditioned medium.Then collecting the cells to detect cell apoptosis ratio by FCM.(3)The cultured mammary cancers cell were divided randomly into 7 groups:the normal control,ADM,MTX,TAX,5-FU,MMC and VCR treatment 1,2,3,4,5,6 group. 1,2,3,4,5,6 group were stimulated by ADM,MTX,TAX,5-FU,MMC and VCR in 5PPC for 48 hours,the normal control group were incubated with equal conditioned medium.Then collecting the cells to detect cell inhibition ratio by MTT.Result:(1)cell morphology and identification.Mammary cancer cell grew in a typical fashion and being adulterated by some of interstitial cells and its characteristics were identified by positive immunostaining of glycoprotein.Infiltrating ductal carcinoma (the example 1-11),infiltrating lobular carcinoma(the example 12-24),papillary carcinoma(the example 25-27),mucinous adenocarcinoma(the example 28-29).(2)Detecting cell apoptosis ratio by FCM:apoptosis were detected in every group on different degrees.The apoptosis ratio were significantly between the treatment groups and the normal group,P<0.05.Infiltrating ductal carcinoma:TAX hypersensitive, ADM 5-FU sensitive,MTX MMC VCR drug fast.infiltrating lobular carcinoma:TAX hypersensitive,ADM VCR sensitive,5-FU MTX MMC drug fast.papillary carcinoma:TAX hypersensitive,ADM 5-FU sensitive,MTX MMC VCR drug fast. mucinous adenocarcinoma:ADM TAX hypersensitive,MMC sensitive,MTX 5-FU VCR drug fast.(3)Detecting cell inhibition ratio by MTT:inhibition were detected in every group on different degrees.The apoptosis ratio were significantly between the treatment groups and the normal group,P<0.05.Infiltrating ductal carcinoma:TAX ADM 5-FU hypersensitive,MTX VCR sensitive,MMC drug fast.infiltrating lobular carcinoma:ADM,TAX,5-FU hypersensitive,MTX,MMC,VCR sensitive.papillary carcinoma:ADM,TAX,5-FU hypersensitive,MTX,VCR sensitive,MMC drug fast.mucinous adenocarcinoma:ADM TAX 5-FU hypersensitive,MMC MTX VCR sensitive.Conclusion:(1)the individual sensitivity of primary culture cell of mammary cancer is a way to prediction chemosensitivities.(2)Because of needing bulk cell,FCM was restriction in some of the example.(3)At present,MTT is a convent and feasible way to detecting chemosensitivities.
