ObjectiveTo research the relationship of homocysteine(Hcy)and gene polymorphisms of its related enzyme(methylenetetrahydrofolate reductase,MTHFR,Methionine synthase,MS)with Alzheimer's disease(AD)in Han peoples of Shan xi province,discuss the possible function of Hcy and its metabolic enzyme's gene in the development of AD and try to supply some theoretical support in the treatment of plasma elevated Hcy level and gene diagnosis and treatment of AD.MethodsThe plasma Hcy level was measured by means of cyclophorase in 136 people of Han peoples (68 patients with AD and 68 healthy controls).Through adopting the methods of polymerase chain reaction-restrictive fragment length polymorphism(PCR- RFLP)to amplificate the directional gene clips of MTHFR,MS,APOE and determine the gene types of amplification productions.Statistics analyzed the experiment data:through t test,the paper compared Hcy level of case group with that of control group;through x~2 test,the paper compared the gene types and allel frequency of the above two groups.Results The plasm Hcy level of AD group(13.92±5.513)umol/L is significantly higher than that(10.83±3.800)umol/L of control group(P=0.001).RR equals to 1.154.95%confidence interval is 1.057-1.26.MTHFR C677T gene frequency is 8.82%,64.71%,26.47%in AD group and 16.18%,42.65%,41.17%in the control group.Generally the gene types vary greatly between the above two groups(P<0.05).The frequency of C/T genetype is remarkably higher in AD group(64.71%)than in the latter(42.65%)(P=0.01).The allel frequency has little difference between the two groups.There was no significant difference of the frequency of MS genetypes and allel between the two.The frequency of AD patients'ApoEε4 allel is significancely higher than the control group' s(P<0.001).ConclusionThere must be some possible relationship between the elevated Hcy level and the development of AD.Gene polymorphism led by gene mutation of MTHFR C677T is possibly involved in the AD's inheritance mechanism.There is no relationship between the MS gene mutation and AD.The elevated Hcy level has something to do with inherited and environmental factors of Han peoples of Shanxi province.Gene treatment,acidum and Vit B treatment can help in the treatment of AD.
